摘要
目的探讨半胱氨酰天冬氨酸特异性蛋白酶(Caspase)-3在乙醇致睾丸损伤中表达的变化,并探讨其意义。方法将24只雄性昆明小鼠随机分为对照组和实验组,每组12只,连续对实验组小鼠腹腔注射乙醇14d,建立睾丸损伤模型,处死所有小鼠并收集睾丸标本,应用实时定量聚合酶链反应(Real—time PCR)和Western blot检测Caspase-3 mRNA和蛋白的表达变化。结果Real-time PCR结果显示实验组Caspase-3 mRNA表达(3.35±0.13)显著高于对照组(P〈0.05);Western blot结果显示Caspase-3蛋白表达(44.51±8.94)显著高于对照组(140.66±6.25,P〈0.05)。结论Caspase-3作为重要的促凋亡基因参与乙醇致睾丸损伤的过程。
Objective To explore the changes and significance of expression of Caspase-3 in the ethanol-induced testis injury model. Methods Twenty-four adult male mice were randomly divided into con- trol (n = 12) and ethanol (n = 12) groups. The mice in the ethanol group were given intra-peritoneal injection (IP) of ethanol at a dose of 3 g ( 15%, v/v) per kg body weight per day during the period of 14 days, to establish testicular damage model. All mice were sacrificed and the testes were collected. Real-time quantitative polymerase chain reaction (Real-time PCR) and Western blotting were applied to detect the expression of Caspase-3 mRNA and protein respectively. Results The results of Real-time quantitative PCR showed that the expression of Caspase-3 mRNA in ethanol group (3. 35 ±0. 13) was significantly higher than in control group (P 〈 0. 05). Western blotting revealed that the Caspase-3 protein expression (44. 51 ± 8.94) was significantly higher in ethanol group than in control group (140. 66 ±6. 25, P 〈0. 05 ). Conclusion Caspase-3 as an important pro-apoptotic gene may participate in the process of testicular damage by ethanol.
