摘要
目的观察促血管生成素-2(Ang-2)在大肠癌组织中的表达及其与临床病理的联系,并探讨其临床意义。方法采用逆转录-聚合酶链反应(RT—PCR)法检测40例大肠癌组织和正常肠黏膜中Ang-2 mRNA的表达,另用Western blot法检测其中10例肿瘤组织和正常组织中Ang-2蛋白的表达;用免疫组织化学法检测其中10例肿瘤组织中Ang-2蛋白的表达。结果60.0%(24/40)的大肠癌组织中Ang-2mRNA强表达,而正常肠黏膜中7.5%(3/40)弱表达,36例阳性表达的肿瘤组织中Ang-2 mRNA表达的平均吸光度值为4.3296±0.2809,而30例正常黏膜上皮的平均吸光度值为3.3097±0.2914,两者差异有统计学意义(P〈0.01);Ang-2蛋白在大肠癌中的表达与肿瘤的病理类型、侵犯层次、淋巴结转移和病理分期有关,且随病程进展表达呈增强趋势。结论Ang-2参与了大肠癌组织新生血管的形成及维持,与大肠癌的进程一致。
Objective To evaluate the expression of angiopoietin-2 (Ang-2) in human colon carcinomas tissues and its relation with clinicopathological parameters in order to clarify the role of Ang-2 in the progression of tumors. Methods Forty cases of colon carcinomas were studied. The relationship between the mRNA expression of Ang-2 and clinicopathological features were examined by reverse transcrip-tion-polymerase chain reaction (RT-PCR) and Western blotting. Immunohistochemical method was used to detect the expression of Ang-2 protein in 10 samples of cancer tissue. Results Ang-2 mRNA expression was significantly detected in 60. 0% of human colon carcinomas tissues and 7.5% of normal tissues, the expression of Ang-2 mRNA in 36 cases of positive expression in tumor tissue, the average absorbance value was 4. 329 6± 0. 280 9, while the average absorbance of 30 normal mucosal epithelia/ values for 3. 309 7 ± 0. 291 4 (P 〈0. 01 ). The Ang-2 expression was related to the pathological types, nodal invasion and lymph node metastasis stage of colon carcinomas. The patients with colon carcinoma strongly positive for Ang-2-expression showed more advanced stages or disease than those weakly positive for Ang-2 expression. Conclusion Ang-2 might be an important factor in tumor angiogenesis and growth in human colon carcinomas.
