摘要 目的:建立激素类药物中氢化泼尼松、倍他米松、醋酸泼尼松、醋酸地塞米松、醋酸氟轻松、氢化可的松、地塞米松、曲安奈德、醋酸泼尼松龙、醋酸可的松、醋酸曲安奈德、氯氟舒松共12个化合物的HPLC快速检测方法。方法:采用Prevail C18(7mm×53 mm,3μm)色谱柱,乙腈-水(43∶57)为流动相,流速2.5 mL·min-1,检测波长224 nm(氢化泼尼松、醋酸泼尼松龙)、240 nm(倍他米松、醋酸泼尼松、醋酸地塞米松、氯氟舒松)、230 nm(醋酸氟轻松)、250 nm(氢化可的松)、248 nm(地塞米松)、237 nm(曲安奈德、醋酸曲安奈德)、245 nm(醋酸可的松);将以上激素类药物分为2组,采用相对容量因子和紫外光谱相似度双指标进行定性,采用相对校正因子法进行定量分析。结果:相对容量因子和紫外光谱相似度定性的准确度均较高,相对校正因子法与外标法定量的结果一致,建立了在10 min内快速检测12个激素类化合物的分析方法。结论:本实验采用相对容量因子和紫外光谱相似度双指标进行12个激素类化合物定性,相对校正因子法定量,使HPLC检测结果更准确,适用于此类化合物的药品的快速监测。 Objective: To establish an HPLC method for the simultaneous fast determination of 12 hormone compounds:hydroprednisone ,betamethasone,prednisone acetate,dexamethasone acetate,fluocinonide,hydrocortisone,dexamethasone,triamcinolone acetonide,prednisolone acetate,cortisone acetate,triamcinolone acetonide acetate halcinonidedcorten. Methods: A Prevail C18 chromatographic column (7 mm×53 mm,3 μm) was used,the mobile phase was acetonitrile-water(43: 57) at a flow rate of 2.5 mL·min^-1,and the detection wavelengths were 224 nm (hydroprednisone,prednisolone acetate),240 nm (betamethasone,prednisone acetate,dexamethasone acetate,halcinonidedcorten),230 nm (fluocinonide),250 nm (hydrocortisone),248 nm (dexamethasone),237 nm (triamcinolone acetonide,triamcinolone acetonide acetate ),and 245 nm (cortisone acetate).The above hormone compounds were divided into two groups,and the relative capacity factor and UV spectrum similarity index were determined for identification of each group of compounds.Relative correction factor method was used for quantitative analysis. Results: The accuracy of the relative capacity factor and UV spectrum similarity index was higher in qualitative study,and the same quantitative results were obtained for the relative correction factor method and the external standard method.The HPLC method for rapid and simultaneous determination of 12 hormone compounds was established,and the detection was completed within 10 minutes. Conclusion: The experiment adopted two indices including the relative capacity factor and the UV spectrum similarity index for qualitative analysis of 12 hormone compounds.The quantitative method using the relative correction factor increased the accuracy of the detection results by HPLC.The method is applicable for the rapid monitoring of such drugs.
