摘要 目的:运用Plackett-Burman实验设计联用正交实验考察各影响因素显著性,筛选出川牛膝多糖制备最佳工艺,为其开发与应用提供依据。方法:运用Plackett-Burman实验设计筛选主要影响因素,以洗脱液浓度、洗脱剂用量、洗脱流速为考察因素,浸膏收率及多糖含量为考察指标,采用综合评分法,利用正交试验优选最佳工艺,并进行验证分析。结果:川牛膝多糖的最佳纯化工艺:S-8大孔树脂用4 BV的0.3 mol/L NaCl溶液以洗脱流速为4 BV/h条件下富集纯化。多糖含量达到90%,浸膏收率为45%。结论:利用Plackett-Burman实验设计联用正交试验确定了川牛膝多糖的纯化工艺,该方法稳定,简便易行。 Objective: To optimize the process of extracting effective constituents from Cyathula officinalis Kuan by Plackett- Burman combined with orthogonal lestmethodology. Methods : Major influence factors used by Plackett-Burman, L9 ( 3^4 ) orthogonal test was applied to analyse the three factors, including concentration of eluent, amount and flowing rate. Results: Optimum process condition was S-8 macroporous resin on the condition of 4BV,0. 3mol/L NaCl as eluting solvent on flowing rate 4BV/h, the polysac- charose and extract yield was more than 90% and 45%. Conclution: The optimum process is simple and more convenient for extrac- ting the polysaccharose from Cyathula officinalis Kuan with an higher precision and predictability.
