电针对脑梗死大鼠患侧海马CA3区GAP-43和c-fos的影响及其与学习记忆的关系 被引量:5

摘要 目的:通过观察电针对脑梗死大鼠患侧海马CA3区GAP-43和c-fos的影响及其与学习记忆的关系,探讨电针对脑梗死大鼠学习记忆能力、中枢神经系统的可塑性影响及其可能机制。方法:56只雄性Wistar大鼠随机选取8只作为假手术组,其余48只脑梗死造模成功的大鼠随机分为脑梗死自由活动组(对照组)和脑梗死电针组(电针组)。电针组于术后24 h开始进行电针针刺"百会","大椎"穴。用水迷宫法检测学习记忆能力,用GAP-43和c-fos试剂盒SABC显色法检测患侧海马CA3区GAP-43和cfos的表达情况。结果:电针组大鼠Morris水迷宫逃避潜伏期短于对照组(P<0.05)。电针组大鼠在平台象限游泳时间占时间之比和经过平台的次数明显大于对照组(P<0.05);海马CA3区GAP-43蛋白阳性表达神经元数在对照组呈现出随着时间的推移而逐渐下降的趋势,而在电针组则呈现为先上升再下降的趋势,在每个时间段上,电针组GAP-43蛋白阳性表达神经元数均比对照组多,该差异具有显著性(P<0.05);电针组c-fos蛋白阳性表达神经元数均比对照组多,该差异具有显著性(P<0.05)。结论:电针"百会","大椎"穴能增加海马CA3区GAP-43和c-fos的表达,使细胞受到保护,细胞凋亡减少,海马最终得以保存更多神经元;明显改善脑缺血大鼠的神经和学习记忆能力。 Objective: This study aims to explore the effects and mechanism of electro acupuncture on the learning and memo- ry, as well as plasticity of central nervous system in rats with cerebral infarction, through observing the effects of electro acupuncture on the learning and memory as well as GAP-d3 and c-fos in the hippocampns neurons of rats with cerebral infarction. Methods: 8 of 56 rats were selected out as sham operated group. The remaining 48 rats were randomly divided into two groups afer occlusion of MCAO : 24 rats in both electro acupuncture group and control group. Electro acupuncture ( "Baihui Dazhui" ) were given to the rats of the Electro acupuncture group 24 hours after operation, while those in control groups were reared in their original living state. Learning and memory was assessed by using Morris water maze. The expression of GAP43 and c-fos in hippocampus was dis- played using immunohistochemistry ( the method was SABC ) . Results: Morris water maze learning ability in electro acupuncture groups exceeded that of control group, and there is obvious difference compared with control group ( P 〈 0. 05 ) . Further more, it is obvious that the percent of swimming time in the platform quadrant and the frequency of cross the platform in electro acupuncture groupthan that in the control group ( P 〈 0. 05 ) . The result of immunohistochemistry indicated that the positive expression of GAP-43 and c- fos in hippoeampus CA3 region at Electro acupuncture group exceeded that of eontrol group ( P 〈 0. 05 ) . Conclusion : The results suggested that the abilities of learning and memory in cerebral infarction rats were affected by Electro acupuncture ( "Baihui Dazhui " ) , which may be accomplished by increasing GAP-43 and c-los expression in hippoeampus CA3 region, then protecting hippoeampus cells from apoptosis. Electro acupuncture ( "Baihui Dazhui " ) can promote recovery of the cerebral infarcted neural function and improve the ability of learning and memory, in cerebral infarction rats .
