乳腺癌组织HR与HER-2和SATB1表达相关性研究 被引量:6

Expression of HR and its correlations with HER-2 and SATB1 in breast cancer tissues

摘要 目的:检测乳腺癌组织中激素受体(hormone receptor,HR)、人表皮生长因子受体2(human epidermal growth factor receptor 2,HER-2)和特异性核基质结合蛋白(special AT rich sequence binding protein 1,SATB1)的表达,探讨HR与HER-2、SATB1及临床病理参数的关系。方法:应用免疫组织化学方法检测乳腺癌患者雌激素受体(estrogen receptor,ER)、孕激素受体(progesterone receptor,PR)、HER-2及SATB1蛋白的表达,荧光原位杂交方法检测HER-2基因扩增状态,统计学方法分析HR(包括ER及PR)的表达与HER-2、SATB1及临床病理参数之间的相关性。结果:乳腺癌组织ER及PR的表达均与患者年龄正相关(r=0.286,P=0.010;r=0.249,P=0.026),ER的表达与肿瘤分级负相关(r=-0.306,P=0.006);ER及PR的表达与肿瘤大小、组织学类型、淋巴转移情况及TNM分期均无明显相关性,P值均>0.05。HR的表达与SATB1、HER-2及SATB1/HER-2双阳性表达均呈负相关关系(r=-0.248,P=0.027;r=-0.392,P<0.001;r=-0.450,P<0.001)。结论:HR阳性患者治疗及预后相对较好;乳腺癌组织HR的表达与HER-2和SATB1呈负相关关系,三者之间可能存在相互联系的信号通路。 OBJECTIVE:To detect the expression of hormone receptor (HR) ,human epidermal growth faclor recep tor 2 (HER 2) and specific nuclear matrix binding protein I(SATB1) in breast cancer,and explore the correlations be tween HR and HEP- 2,SATB1 as well as the elinicopathological features. METHODS: Immunohistoehemistry was used to detect the expression of estrogen receptor (ER), progesterone receptor (PR), HER 2 and SATB1 protein in breast cancer patients. FISH was used to test the amplification of HER-2 gene. Correlations of HR with HER-2, SATB1 and clinicopathologic features were analyzed. RESULTS: The expression of ER, PR was positively correlated with patient age in breast cancer(r=0. 286, P= 0. 010;r= 0. 249,P 0. 026), while ER expression was negatively correlated with tumor grade(r=-0. 306, P = 0. 006). No correlations were found between ER or PR expression and tumour size, pathological type,lymph node status as well as TNM stage (All P〈0.05). HR expression was negatively correlated with HER-2, SATB1 expression and SATB1/HER-2 co-expression(r= 0.248,P=0.027;r=-0.392,P〈0.001;r=-0.450,P〈 0. 001). CONCLUSIONS: HR positive expression may indicate a better therapitic and prognostic outcome. Negative correlations existed between the expression of HR and SATB1/HER 2 in breast cancer tissues; and the phenomenon possibly suggested their cross-talks though the signaling pathways.
