摘要 以牛血清白蛋白(BSA)为酶制剂模型,用异硫氰酸荧光素(FITC)标记BSA制备了荧光蛋白质FITC-BSA。测试表明:FITC-BSA的等电点及分子质量与BSA基本相同。用FITC-BSA处理脱灰裸皮后,通过荧光显微镜能直观地观测到FITC-BSA在皮内的传递和分布情况。这些结果表明:采用本研究建立的荧光示踪技术,可以准确地检测蛋白质类材料在皮内的传质行为,该技术适用于研究制革过程酶的传质规律及作用机理。 The mass transfer and action mechanism of enzymes in leather processing remained unclear due to the lack of an ap- propriate method of effectively observing the transfer of enzymes in skin/hide, which had restricted the wide use of enzymes in leather industry. The bovine serum albumin(BSA) was employed as a model of enzyme preparations and labeled by using fluoresce- in isothiocyanate (FITC). The results show that isoelectric point and molecular weight of FITC - labeled BSA ( FITC - BSA) are similar to those of BSA. The transfer and distribution of FITC - BSA in delimed pelts can be directly observed by fluorescence mi- croscope after the pelts are treated by FITC - BSA. It confirms that the transfer of protein materials in skin/hide can be accurately detected by the fluorescence tracing, and the tracer technique is suitable to investigate the mass transfer and action mechanism of enzymes in leather processing.
