摘要
目的研究天麻多糖对小鼠肝癌H22细胞增殖的影响及其对H22荷瘤小鼠肿瘤生长的抑制作用。方法 MTT法检测不同质量浓度的天麻多糖对小鼠肝癌H22细胞体外增殖的影响;健康小鼠和接种H22瘤株造模昆明种小鼠分成7组(n=12):对照组(0.9%NaCl溶液2 ml),模型组(0.9%NaCl溶液2 ml),环磷酰胺(20 mg·kg-1)组,天麻多糖低、中、高剂量(30、60、90 mg·kg-1)组,观察天麻多糖对H22荷瘤小鼠的抑瘤作用。结果天麻多糖对肝癌H22细胞增殖有显著的抑制作用。且在小鼠体内能明显抑制对H22肿瘤瘤体生长,高剂量(90 mg·kg-1)效果明显(P<0.05);环磷酰胺和天麻多糖合用可使环磷酰胺对白细胞计数、胸腺指数和脾指数的影响降低。结论天麻多糖能抑制肝癌H22细胞的增殖,与环磷酰胺联用可降低环磷酰胺对免疫系统的伤害。
In this study, we aimed to investigate the antitumor effects of polysaccharides from Gastrodia Elata B1 (PGE) on mice bearing H22 solid tumor. Firstly, we stimulated H22 cell with PGE at concentrations of 15, 30, 60, 120, 240 mg .ml^-1 respectively and detected the effect of PGE on cells proliferation by MTT. Then the antitumor effect of different dose of PGE in vivo was observed in mice inoculated subcutaneously with H22 cells, which were equally divided into model group, cyclophosphamide group (CTX, 20 mg/kg), low-, moderate-, and high- dose (30,60 and 90 mg.kg^-1, respectively) PGE group and CTX (20 mg.kg^-1) combined with PGE (60 mg.kg^-1) group, withhealthy mice as control. The result showed PGE, especially at a dose of 60 or120 mg.L^-1, had remarkable inhibitory effect on the proliferation of H22 hepatoma cells in vitro. Furthermore, PGE at a dose of 90 mg.kg^-1 suppressed the tumor growth in vivo. The administration of CTX combined PGE could significantly inhibit the depressant effect of CTX on white blood cell count, thymus index, and spleen index. In conclusion, PGE can inhibit the proliferation of H22 cells, and reduce the damage of CTX on the immune system when combining administration with CTX.
