蜣螂抗良性前列腺增生症活性部位的筛选(Ⅱ) 被引量:4

Screening of active fraction of anti-benign prostatic hyperplasia from Catharsisus molossus Linnaeus( II)

摘要 目的:探讨蜣螂乙醇提取物抗大鼠前列腺增生的影响及其可能机制。方法:去势雄性大鼠皮下注射丙酸睾酮造成良性前列腺增生模型,将模型大鼠分为3组,分别灌胃蜣螂提取液、非那雄胺水溶液和同体积蒸馏水,正常对照组和模型组给予等体积蒸馏水,每天给药一次,连续给药4周,末次给药1 h后,处死大鼠,检测大鼠血清睾酮、雌二醇水平与前列腺指数,观察前列腺组织形态,采用免疫组化法检测表皮生长因子(EGF)、转化生长因子-β1(TGF-β1)在大鼠前列腺组织中的表达情况。结果:蜣螂20g生药/kg使造模大鼠血清睾酮水平增加、雌二醇水平降低;同时使大鼠的前列腺湿重、前列腺指数降低,前列腺组织腺上皮及间质面积缩小;并能降低EGF在前列腺组织中的表达,增加TGF-β1的表达。结论:蜣螂乙醇提取物可能通过抑制前列腺组织中睾酮向二氢睾酮转化,调节大鼠雌/雄激素比例平衡,抑制前列腺上皮组织与间质增生,调节EGF与TGF-β1在前列腺组织中的表达,从而抑制前列腺增生。 Objective: To study the inhibiting effects of theethanol extract from Catharsisus molossus Linnaeus on model rats with benign prostate hyperplasia(BPH) and explore its possible mechanisms. Methods: Model rats with BPH were established by subcutaneously injection of tes- tosterone propionate. The model rats were divided into 3 groups, respectively were fed with the extract from Catharsisus molossus Linnaeus, finasteride solution and the same volume of distilled water. The blank control group and model group were fed with the same solution. Every group was given daily for four weeks. After one hour from last administration, rats were sacrificed. The serum testosterone, ostradiol levels and the prostate index were tested and histomorphology changes of prostate gland were observed. The expression of epidermal growth factor (EGF) and transforming growth factor β1 (TGF-β1) in prostate tissues were detected by immunohistochemical analysis. Results: The extract (20 g crude drug/kg) from Catharsisus molossus Linnaeus increased the level of serum testosterone and decreased estradiol levels of model rats. The prostate weight and prostate index were lowered, while the area of prostate gland epithelial cen and stromal cell were reduced. The extract also reduced expression of EGF and increased expression of TGF-β1 in prostate tissues. Conclusion: The ethanol extract from Catharsisus molossus Linnaeus may inhibit experimental prostatic hyperplasia by suppressing transformation of testosterone into dihydrotestosterone, regulation of estrogen/androgen ratio and expression of EGF and TGF-β1, inhibition of prostate epithelial and stromal hy- perplasia.
