摘要
目的:研究保妇康对慢性盆腔炎大鼠子宫粘膜细胞间黏附分子1(ICMI-1)和转化生长因子β1((TGF-β1)mRNA表达及血清炎性细胞因子含量的影响,以探讨其作用机制。方法:将雌性大鼠随机分为7组,除正常组和假手术组外,其他各组采用混合细菌加机械损伤法建立慢性盆腔炎大鼠模型。保妇康高、中、低剂量组分别以2.07,4.14,8.28g/kg ig给药,连续给药4周,采用半定量荧光免疫技术(RT-PCR)法测定大鼠子宫组织ICMI-1和TGF-β1mRNA的表达,用ELASA法测定血清TNF-α和IL-6的含量。结果:与正常组相比,模型组大鼠子宫黏膜炎性细胞浸润明显,子宫组织ICMI-1(2.03±2.76)和TGF-β1(9.03±2.76)mRNA的表达均明显升高(P〈0.05)。与模型组比较,保妇康2.07~8.28g/kg剂量范围内,均可明显减轻子宫粘膜炎症反应,高剂量组(8.28g/kg)、中剂量组(4.14g/kg)ICMI-1和TGF-β1mRNA的表达均明显降低,其中,高剂量组TGF-β1mRNA(4.11±1.29)的表达下降最明显;低剂量组(2.07g/kg)ICMI-1和TGF-β1mRNA的降低没有显著性差异。高、中剂量组(4.14~8.28g/kg)的IL-6和TNF-α明显降低。结论:保妇康能降低血清炎性细胞因子的含量,下调子宫组织ICMI-1和TGF-β1mRNA的表达,减轻盆腔组织粘连,这可能是其防治慢性盆腔炎的机制之一。
Objective: To study Effect of Baofukang on expression of ICMI-1 ,TGF-β1 mRNA in uterus and inflammatory cytokines in blood serum of chronic pelvic inflammation rats, and to explore possible mechanism of the therapeutic effects. Method: Female rats were randomly devided into 7 groups,in which chronic pelvic inflammation model was made by injection of a mixture of bacteriums suspension into injured uterus except the normal group and sham operation group. Baofukang(2.07, 4.14, 8.28g/kg/d ig)were administrated for 4 weeks. Expression of IC- MI-1 and TGF-β1 mRNA of ophoron and uterus in rats dectectod by RT-PCR. Histopathologic changes of uterus were observed by microscope. IL-6 and TNF-a were measured by enzyme-linked immunosorhent assay (ELISA). Result:In compare with normal group, content of ICMI-1 (2.03 ± 2.76) and TGF-β1 (9.03 ± 2.76) mRNA in model group were rlsed obviously( P 〈 0. 05 ). after Baofukang administrated, Hemorheology of rats was improved obviously in all groups( 2.07± 8.28g/kg). the expression of ICMI-I and TGF-131 mRNA in high(8.28g/kg) and middle(4. 14g/kg) group were lowed severally( P 〈 0.05 ). however, there was no statistically significant difference between low (2.07g/kg) and model group( P 〉 0.05 ). Content of IL-6 and TNF-ctwere decreased obviously in high and middle groups (4.14 - 8.28g/kg). Conclu- sion: The it can prevent and treat chronic pelvic inflammation by decreasing the expression of ICMI-1 and TGF-β1mRNA in ophoron and ute- rus of chronic pelvic inflammation rats and relieving pelvic adhesions.
