摘要
目的:研究双藤痹痛凝胶膏剂对胶原诱导性关节炎(CIA)大鼠的作用。方法:将48只大鼠随机分为正常对照组、空白凝胶膏剂组、阳性组伤湿止痛膏及双藤痹痛凝胶膏剂高、中、低剂量组(3cm×3cm、2cm×3cm、1cm×3cm)。除正常对照组外,其余各组采用尾根部皮下皮下注射胶原乳液免疫建立CIA模型;观察各组大鼠一般情况、体重、关节指数、足趾肿胀度及关节组织的病理学变化。结果:造模后,模型大鼠的足趾肿胀明显,与正常对照组比较,有显著性差异;初次免疫后第28天,双藤痹痛凝胶膏剂3cm×3cm、2cm×3cm、1cm×3cm各组与空白凝胶膏剂组比较,关节指数、足趾肿胀度均降低,有统计学意义;HE病理切片显示,双藤痹痛凝胶膏剂3cm×3cm、2cm×3cm剂量下可抑制CIA大鼠关节组织病理学改变。结论:成功建立CIA大鼠模型;双藤痹痛凝胶膏剂能够抑制CIA模型大鼠足趾肿胀和关节组织病理学改变,降低其关节炎指数。
Objective: To investigate the effect of Shuangteng Bitong cataplasm on collagen-induced arthritis(CIA) rats. Methods: 48 Wistar rats were randomly divided into control group, model group, positive, Shuangteng Bitong eataplasm high, medial and low-dose groups (3cm × 3cm ,2cm × 3cm, 1 cm × 3cm). Except the normal group, collagen emulsion was injected into the tail root of each rat via intradermal injection to induce arthritis. Then, the condition of the rats, body weight, arthritis index, paw edema, histopathological changes were observed in the six groups. Results: In model group,the paw edema increased significantly in comparison with that of rats in the normal group( P 〈 0.01 ). On the 28th day after primary immunization, the arthritis index and paw edema of rats in Shuangteng Bitong cataplasm high, medial and low- dose groups ( 3cm × 3cm ,2cm × 3cm, 1 cm × 3cm) were decreased compared with that of CIA model group ( P 〈 0.05 ). The results of HE staining showed that pathological changes of rats in the Shuangteng Bitong cataplasm of high and medial dose (3cm × 3cm ,2cm × 3cm) had significantly reduced compared with that of rats in the CIA model group. Conclusion : The CIA rat model of RA was successfully established in this study, and the Shuangteng Bitong cataplasm could reduce arthritis index, paw edema and pathological changes of CIA rats.
