清风胶囊对反复吗啡依赖和戒断大鼠焦虑行为及脑内单胺神经递质水平的影响

Effect of Qingfeng capsule on anxiety and intracephalic monoamines neurotransmitter in rats of repeated dependency and withdrawal of morphine

目的:探讨清风胶囊对反复吗啡依赖和戒断引起的大鼠焦虑行为和脑内单胺递质的影响。方法:复制吗啡依赖大鼠成瘾和催促戒断后焦虑模型。将大鼠随机分空白对照组、吗啡模型组、清风胶囊低、中、高剂量组和阳性药地西泮对照组。清风胶囊低、中、高组给药剂量分别为0.5,1.0,2.0 g/kg,连续给药5天。观察动物焦虑行为的改变,并采用荧光分光光度计检测受试大鼠下丘脑单胺类神经递质去甲肾上腺素(NE)、5-羟色胺(5-HT)和多巴胺(DA)的含量。结果:清风胶囊0.5g/kg,1.0g/kg,2.0 g/kg三剂量均能明显缓解吗啡依赖大鼠由于反复躯体依赖和戒断所导致的焦虑行为,并显著减少大鼠下丘脑单胺类神经递质NE、5-HT和DA的含量。清风胶囊0.5,1.0,2.0 g/kg三剂量对大鼠焦虑行为的抑制作用与地西泮比较无明显差异,三剂量组之间亦无明显差异。结论:清风胶囊0.5g/kg,1.0g/kg,2.0 g/kg三剂量均对反复吗啡依赖大鼠戒断后的焦虑行为有明显的治疗效果。

Objective： To study effect of Qingfeng capsule on anxiety and intracephalic monoamines neurotransmitter in rats of repeated dependency and withdrawal of morphine. Methods： Rats model repeated dependency and withdrawal of morphine by dampness was made. The rats were divided randomly into six groups： the control group,the model group,and three groups administered with Qingfeng capsule at low,mediate,and high doses respectively（ 0. 5,1. 0,2. 0 g /kg）,and successively administrated for 5 days. The anxiety behavioristics changes of rats were observed and the content of hypothalamic monoamines neurotransmitter NE,5-HT,DA was recorded with fluorospectro photometer. Results： Three groups of 0. 5,1. 0,2. 0 g /kg of Qingfeng capsule can obviously improve anxiety behavioristics on rats of repeated dependency and withdrawal of morphine,and the level of NE,5-HT,DA was obviously decreased. Inhibitory effect of Qingfeng capsule on rats with anxiety behavioristics was indistinctive difference,compared with diazepam. Three groups of 0. 5,1. 0,2. 0 g /kg of Qingfeng capsule were indistinctive difference（ P ＆gt; 0. 05）. Conclusion： Three groups of 0. 5,1. 0,2. 0 g /kg of Qingfeng capsule had therapeutic effect on anxiety behavioristics of rats with repeated dependency and withdrawal of morphine.
