显微分光光度法测定氮氧化物

摘要 在还原底物NADPH及硝酸盐还原酶的催化下 ,显微分光光度法测定细胞及其培养液的氮氧化物含量 ,得到满意结果 :Sander灵敏度为 3 8× 10 - 6μmol/cm2 ,变异系数CV 3 96% (n =8) ,标准添加回收率 10 0 0 %± 4 96% ,浓度 (C)与吸光值 (A)的线性范围为 0 15~ 1 0 0μmol/L ,其回归方程为C =- 0 0 18+1 0 99A (r =0 9958,P <0 0 1) ,在合适的还原条件下 ,酶促的还原效率 95 9%。本法适用于生物样品中微量氮氧化物的分析。 A method was developed for the determination of nitric oxides in cells and its culture medium by microspectrophotometry with NADPH and catalyzing of nitrate reductase(NR).As the results,it was found that the Sander sensibility is 3 8×10 -6 μmol/cm 2,the coefficient of variation is 3 96%( n =8),the recovery of using the standard addition method is (100±4 96)%.A linear calibration curve was obtained in the absorbance( A )~concentration range of (0 15~1 00)μmol/L,and the regression equatoin C =-0 018+1 99 A(r =0 9958, P <0 01).The reduction efficiency of NO - 3 in enzyme catalysis was 95 9%.The present method is good enough for quantitative analysis of nitric oxides in biological samples.
