毛细管电泳和质谱联用在线预浓缩酚酸类化合物及其抗氧化活性评价 被引量:3

Analysis of Phenolic Acids and Their Antioxidant Activity by Capillary Electrophoresis-mass Spectrometry with Field-amplified Sample Injection

摘要 目的建立一种新型高效的毛细管电泳和质谱联用(CE—MS)在线预浓缩方法,用于检测丹参样品中酚酸类化合物中4种亲水活性成分(丹参素、丹酚酸B、咖啡酸和紫草酸),并对其抗氧化活性作出评价。方法利用CE—MS在线预浓缩技术,采用未涂层石英毛细管柱(90cmx50μm),电压:27kV。样品的进样压力:50mhar。选样时间:3~300S,毛细管温度:25℃。鞘液:5mmol·L-1的乙酸氨溶液(甲醇溶解),流速:4mL·min-1。结果该技术灵敏度比通常的流体注射提高了30~50倍。结论该方法可用于分析丹参药材中丹参素、丹酚酸、咖啡酸、紫草酸等酚酸类化合物,也可以用于评价丹参样品中酚酸类化合物的抗氧化成分。 Objective A capillary electrophoresis-mass spectrometry (CE-MS) method with field-amplified sample stacking was developed to analyze and detect four hydrophilic active components(danshensu, salvianolic acid B, caffeic acid and lithospermic acid) presented in Radix Salviae Miltiorrhizae samples, and to evaluate their antioxidant activity. Methods The assay was performed on an uncoated fused-silica capillary of 95 cm x 50 μm, and a positive voltage of 27 kv was applied. The sample was injected under pressure of 50 mbar for 3-300 s and the temperature of capillary was kept at 25 ℃. Sheath liquid consisted of 5 mmol ·L-1 ammonium acetate in methanol solution, flowing at 4 mL·min-1. Results By utilizing the preconcentration technique, the sensitivity of CE-MS was enhanced by approximately 30-50 folds in terms of detection limit compared to common hydrodynamic injection. Conclusion The established method can be used in the analysis of components (danshensu, salvianolic acid B, caffeic acid and lithospermic phenolic acid in Radix Salviae Miltiorrhizae, and can also be applied to evaluate the antioxidant activity of
