过表达相关蛋白对酿酒酵母利用半乳糖的影响 被引量:1

Effects of Over-expression of Some Enzymes on Galactose Utilization of Saccharomyces Cerevisiae

摘要 以PGK1为启动子,以YEp352为载体,构建了SEC53和PGM2基因表达载体,获得酵母转化子AYSEC和AYPGM。通过对比重组菌株与亲本的发酵性能,研究了过表达SEC53、PGM2基因对酿酒酵母半乳糖代谢的影响。结果表明,过表达SEC53、PGM2基因可显著提高酿酒酵母利用半乳糖的速度;在半乳糖培养基中,重组菌株AYSEC和AYPGM的总发酵时间分别缩短了37.5%和25%;在混合糖培养基中,重组菌株在一定程度上降低了葡萄糖的阻遏效应,AYSEC和AYPGM总发酵时间分别缩短了28.6%和14.3%。 The recombined vectors were constructed to overexpress SEC53 and PGM2 genes by using PGK1 as the promoter and plasmid YEp352 as the carrier. Then recombinant strains AYSEC and AYPGM were obtained. The effects of the over-expression of SEC53 and PGM2 on galactose utilization in S.cerevisiae strain were investigated through comparing the fermentation performance of parent strains and mutant strains.The results showed that, the over-expression of SEC53 and PCM2 genes could significantly improve galactose utilization rate. In glalactose culture mediums, AYSEC and AYPGM could shorten total fermentation time by 37.5 % and 25 %, respectively. In glucose-galactose culture mediums, the recombinant strains could weaken the glucose repression and shorten the fermentation time by 28.6 % (AYSEC) and 14.3 % (AYPGM), respectively.
