MBP刺激的PBMC培养上清对内皮细胞促进T细胞增殖的作用

摘要 目的探讨髓磷脂碱性蛋白MBP刺激的PBMC培养上清作用的人脐静脉内皮细胞HUVEC对T细胞增殖的影响。方法用细胞免疫化学染色和FCM检测HUVEC的VCAM1ICAM1和HLADR的表达用MTT法测定MBP刺激的PBMC对T细胞增殖的影响。结果与对照相比较MBP刺激的PBMC培养上清可诱导HUVEC特异表达VCAM1和HLADR上调ICAM1的表达。经MBP作用的HUVEC可明显促进T细胞的增殖。结论MBP刺激PBMC的培养上清可引起内皮细胞结构和功能改变促进T细胞增殖。 Aim To explore the effect of human umbilical vein endothelial cell(HUVEC) co incubated with the culture supernatant of MBP stimulated PBMC on the T cell proliferation. Methods The expressions of VCAM 1,ICAM 1and HLA DR on HUVEC were examined by immunocytochemical staining and FCM. The T cell proliferation was tested by MTT assay. Results The culture supernatant of MBP stimulated PBMC could induce the specific expression of VCAM 1 and HLA DR and upregulate expression of ICAM 1 on HUVEC compared with control. The HUVEC co incubated with the culture supernatant could evidently promote T cell proliferation. Conclusion The culture supernatant of MBP stimulated PBMC can give rise to changes of HUVEC structure and function, and promote T cell proliferation.
