皮下蜜蜂毒致持续性自发痛反应的两种定量方法及吗啡抑制效果(英文) 被引量:3

摘要 目的 比较两种定量检测皮下蜜蜂毒所致自发痛反应的方法及吗啡的镇痛效果 .方法 采用大鼠足底皮下注入蜜蜂毒致痛模型 ,分别以自发缩足反射次数和四级负重记分两种定量方法观察记录伤害性反应 .结果 静脉吗啡预处理对大鼠足底注入蜜蜂毒致自发痛反应具有一定抑制作用 ;以自发缩足反射次数定量 ,6种剂量 (0 .0 15 ,0 .15 ,0 .3,0 .47,1.5和 3.0 mg·kg- 1 )吗啡呈剂量依赖性抑制效应 ,抑制率分别为 :17,39,48,5 2 ,6 2和 89% ,与对照组比较均具有统计学差异 (P<0 .0 5 ) ,其半数抑制有效量 (ED5 0 )为 0 .2 9mg·kg- 1 ;而采用四级负重记分法定量 ,仅吗啡 1.5 mg· kg- 1实验组的抑制具有统计学意义 (P<0 .0 5 ) .结论 吗啡对大鼠足底皮下注入蜜蜂毒致自发缩足反射次数具有剂量依赖性抑制作用 .对于蜜蜂毒致痛模型 ,自发缩足反射次数做为自发痛反应定量指标较四级负重记分法简便、稳定。 AIM To compare two methods of quantitating spontaneous pain related responses on evaluating the analgesic effect of morphine. METHODS After subcutaneous injection of bee venom into the plantar of one hindpaw in rats, the nociceptive responses were evaluated by two different methods: by counting the number of flinching reflex and by the four grade weight scoring. RESULTS The suppressive effect of pretreatment with morphine i.v. was found. Pretreatment with 0.015, 0.15, 0.3, 0.47, 1.5, 3.0 mg·kg -1 morpine i.v. significantly produced a dose dependent suppression of the bee venom induced spontaneous pain related responses ( P <0.05). The inhibitory rates were 17, 39, 48, 52, 62 and 89 per cent. The ED 50 of morphine was 0.29 mg·kg -1 . Pretreatment with 1.5 mg·kg -1 morphine i.v. produced significant suppression on the bee venom induced lifting/licking nociceptive score. CONCLUSION Pretreatment with morphine i.v. can produce a dose dependent suppression on the bee venom induced spontaneous flinching reflex. In the bee venom test, the spontaneous flinching reflex as quantitative measure of spontaneous pain related responses is more simple, stable and objective than lifting/licking nociceptive score.
