摘要
目的探讨补肾活络方治疗多囊卵巢综合征(PCOS)的药理机制。方法 60只雌性SD大鼠分为空白组、模型组、阳性药物组、补肾活络方高、中、低剂量组,每组10只。采用胰岛素联合绒毛膜促性腺激素(HCG)造成的多囊卵巢(PCO)大鼠模型,以补肾活络方高、中、低3个剂量以及达英-35分别作用于PCO模型大鼠12 d后,放免法检测动物血清促黄体生成素(LH)、睾酮(T)、促卵泡生成素(FSH)、雌二醇(E2)的含量,光镜下观察各组大鼠卵巢形态。结果与模型组相比,补肾活络方高、中剂量组大鼠卵巢重量显著减轻(P<0.05),卵泡数目显著减少(P<0.05),组织形态明显改善,血清T、LH含量显著降低(P<0.05)。结论补肾活络方可能通过降低T、LH的水平而达到促进卵泡发育和排卵的作用。
objective To investigate the pharmacologic mechanism of Bushen Huoluo Fang in the treatment of polycystic ovary syndrome (PCOS). Methods Female SD rats (n = 60 ) were randomly divided into blank group, model group, positive drug group, and high-dose, mid-dose and low-dose Bushen Huoluo Fang groups (high-dose, mid-dose and low-dose groups, each n = l0 ). The model of PCOS was established by using insulin combining human chorionic gonadotropin (HCG). Bushen Huoluo Fang (in three doses) and Diane-35 were administrated in corresponding groups respectively for 12 d, and then the levels of serum luteinizing hormone (LH), testosterone (T), follicle-stimulating hormone (FSH) and estradiol ( E2 ) were detected by using radioimmunoassay, and the changes of ovarian morphology were observed with light microscope. Results Compared with model group, the weight of ovary (P 〈 O. 05 ), follicle amount ( P 〈 0.05 ) and levels of serum T and LH ( P 〈 0.05 ) decreased significantly, and histomorphology changes were improved in high-dose and mid-dose groups. Conclusion Bushen Huoluo Fang can promote follicular development and ovulation through reducing the levels of T and LH.
