摘要
目的 探讨降低黏着斑激酶 (FAK)的表达对FAK高表达的人肝癌细胞株SMMC 772 1恶性生物学行为影响。方法 用Western杂交法比较不同肝癌细胞株FAK含量的差别 ,构建FAK反义质粒 ,转染至FAK高表达的细胞株 ,研究其多种细胞生物学行为的变化。结果 SMMC 772 1FAK含量比正常人肝细胞L0 2明显增高 ,转染FAK反义质粒后 ,SMMC 772 1细胞生长受到抑制 ,软琼脂培养克隆形成率下降 ;细胞周期分析 ,S期细胞下降 15 % ;黏附能力下降 ;细胞表面整连蛋白含量不变。结论 在SMMC 772 1中存在FAK过表达 ,降低FAK表达能部分逆转该肝癌细胞株的恶性表型。
Objective To study the effect of reducing FAK expression on the malignant biological behaviors of FAKoverexpressed hepatocellular carcinoma cell line SMMC7721. Methods[WT5”BZ] FAK expression of different hepatocelluar carcinoma cell lines was detected by Western blot analysis.Antisense plasmid of FAK was constructed and transfected into SMMC7721. Results SMMC7721 expressed much more FAK than did hepatocyte line L02. When transfected with autisense FAK, it showed reduced growth rate and colony formation in soft agar, and the number of cells in S phase was reduced by 15%. The adhesion ability of antisense FAKtransfeected cells was also decreased with no change of integrin expression. Conclusion FAK is overexpressed in the human hepatocellular carcinoma cell line SMMC7721. Reducing the FAK expression can partly reverse the malignant behaviors of the cell line.
