摘要
目的通过观察多囊卵巢综合征(PCOS)大鼠子宫内膜葡萄糖转运蛋白-4(GLUT-4)和胰岛素样生长因子(IGF-1)的表达以及应用二甲双胍后子宫内膜GLUT-4和IGF-1表达的改变。探讨影响PCOS大鼠子宫内膜容受性可能的机制。方法选取6周龄清洁级雌性Wistar大鼠70只,按体质量计算分为对照组20只、模型组50只。应用来曲唑灌胃建立PCOS大鼠模型,造模成功后,模型组抽取40只分取二甲双胍治疗组20只和PCOS组20只分别喂服二甲双胍和0.9%氯化钠注射液。14d后麻醉大鼠并取材,光镜观察子宫内膜的情况,免疫组织化学法测定3组子宫内膜GLUT-4和IGF-1的表达水平。结果 PCOS组大鼠子宫内膜腺上皮和间质中GLUT-4蛋白表达明显低于对照组(P<0.05),IGF-1的表达明显高于对照组(P<0.05)。治疗组GLUT-4表达较PCOS组显著增高(P<0.05)但仍低于对照组(P>0.05),治疗组IGF-1表达比PCOS组降低(P<0.05)与对照组相比仍然较高(P<0.05)。结论 PCOS大鼠子宫内膜GLUT-4和IGF-1均与对照组有明显的差异。通过二甲双胍治疗后GLUT-4和IGF-1的变化明显接近正常对照组,说明GLUT-4和IGF-1与PCOS子宫内膜改变有相关性。
Objective To observe the expression of glucose transporter proteins 4(GLUT-4)and insulinlike growth factor-1(IGF-1)of endometrium in rats with polycystic ovarian syndrome(PCOS),and the change of GLUT-4and IGF-1of endometrium in rats after therapy with metformin.To investigate the mechanisms affecting endometrial receptivity of rats with PCOS.Methods To set up an animal model by using the non-steroidal aromatase inhibitor-letrozole.Seventy female Wistar rats of six weeks age were randomly divided into three groups:the control group(n=20),PCOS model group(n=20),metformin treatment group(n=20).The control group were feed normally.The latter 2groups were treated by metformin and normal saline,respectively.After 14days of treatment,the rats were anesthetized and sacrificed.Endometrial condition was observed by light microscope.Using the immunohistochemical staining technique the expression level of GLUT-4and IGF-1was detected.Results The expression of glucose transporter proteins 4(GLUT-4)proteins of endometrial in PCOS rats was significantly lower(P〈0.05)than that in control group,and the expression of(IGF-1)protein in PCOS rats was higher than that in control group(P〈0.05).The expression of GLUT-4in the treatment group increased(P〈0.05),but was still lower than that in control group(P〉0.05).However,compared with PCOS group,the expression of IGF-1protein in treatment group was decreased(P〈0.05),but was still higher than that in control group(P〈0.05).Conclusion Compared with control group,GLUT-4and IGF-1of endometrium in PCOS group were obviously different.Through the metformin treatment may reverse the situation.It is concluded that GLUT-4and IGF-1have correlation with change of PCOS endometrial.
