摘要
目的:观察补肾益气方对自然衰老大鼠皮质酮及IL-2基因表达的影响。方法:将20月龄老年大鼠随机分为对照组与补肾组,各10只。另设3月龄青年组。其中老年补肾组自分组日起以饮料方式饲喂补肾益气方药液,连续3个月,其余两组予正常饮水和饲料。于干预3月结束时,取腹主动脉血和脾脏。采用ELISA法检测大鼠血清IL-2含量;EIA法检测大鼠血清皮质酮含量;qRT-PCR法检测大鼠脾淋巴细胞中IL-2、糖皮质激素受体(GR)mRNA的表达;Western blotting法检测大鼠脾淋巴细胞GR蛋白表达。结果:与青年组相比,老年对照组大鼠血清皮质酮含量明显升高(P<0.05),血清IL-2含量明显降低(P<0.05),脾淋巴细胞IL-2 mRNA、GR mRNA和蛋白表达明显下降(P<0.05);与老年对照组比较,老年补肾组血清IL-2水平明显升高(P<0.05),GR mRNA和蛋白表达明显升高(P<0.05)。结论:补肾益气方可通过调节老年大鼠异常的皮质酮/糖皮质激素受体通路,改善IL-2基因表达的增龄性衰退。
Objective: To investigate the effect of " Bushen Yiqi Formula" on expression of IL-2 and secretion of endogenous corticosterone in senescent rats. Methods: Twenty months rats were randomized into control group and kidney- replenishing group, with 10 rats ineach group Young group was established, Kidney-replenishing group was treated with "Bushen Yiqi Formula" for 3 months continuously, and the others were treated with water and fodder. The content of serum IL-2 was detected by ELISA and the corticosterone content was detected by EIA; the mRNA expressions of IL-2 and GR in splenic lymphocytes were detected by qRT-PCR; the expression of glucocorticoid receptors (GR) protein was detected by Western blotting. Results: Compared with young group, the corticosterone content in aged control group was increased markedly (P 〈 0.05 ), but the IL-2 mRNA expression in splenic lymphocytes, serum IL-2 and GR expression were decreased markedly ( P 〈 0.05 ) ; Compared with aged control group, the expressions of IL- 2 and GR were improved in aged kidney- replenishing group, and the content of corticosterone was down-regulated. Conclusion: "Bushen Yiqi Formula" can improve the IL-2 expression by regulating corticosterone/GR pathway in aged rats.
