应用AB基因分析仪构建犬个体识别荧光标记检测方法

摘要 选择16个STR基因座,应用五色荧光标记技术,使用ABI3130基因分析仪对80头拉布拉多犬的遗传多态性进行检测分析。研究表明:这16个STR基因座建立的荧光标记复合扩增体系,各个基因座扩增平衡,结果均一稳定;16个STR基因座共检测到164个等位基因,平均杂合度(H)和平均多态信息含量(PIC)分别为0.881和0.813;而累积非父排除率(CPE)和累积个体识别率(TDP)分别为0.999999999997和0.9999999999999999。这16个STR基因座建立的荧光标记复合扩增体系,可以有效用于犬的个体识别和亲权鉴定。 We tried to construct a five-fluorescent-labeled multiplex amplification system including 16 STR loci ,analyze the genetic polymorphisms of 80 Labrador Retrievers by ABI3130 genetic analyzer .The results showed the 16 STR loci multiplex system was constructed successfully ,with good amplification product ,good intracolor and intercolor balance .164 alleles were detected in 16 STR loci ,the average PIC and heterozygosity were 0 .881 and 0 .813 respeclively .The cumulative probability of paternity exclusion (CPE) and total power of discrimination (TDP) were 0 .999999999997 and 0 .9999999999999999 respec-tively .The five-fluorescent-labeled 16 STR loci multiplex system had been successfully constructed ,could be applied to canine individual discrimination and paternity identification .
