紫玉兰色素超声结合酶法提取工艺及其稳定性研究 被引量:5

Study on the Optimization of Ultrasonic-assisted Enzymatic Extraction of Pigment from Magnolia Liliiflora and its Stability

摘要 本文以紫玉兰为材料,采用超声波结合酶法提取紫玉兰色素,采用单因素和正交实验研究不同酶用量、酶解时间、酶解温度、乙醇体积分数、超声时间、料液比等条件对色素提取的影响,并确定最佳的提取工艺。同时研究光照、pH值、不同浓度NaCl、碳水化合物和金属离子对紫玉兰色素稳定性的影响。结果表明:紫玉兰色素提取的最佳工艺参数为纤维素酶用量4mg/g(花瓣湿重),按1∶15的料液比加入60%的乙醇溶液,在50℃酶解80min,超声辅助提取10min。该色素对强碱和光照不稳定,NaCl和碳水化合物对其有护色作用,Fe3+和Cu2+离子对色素稳定性影响较大。 In this paper ,the pigment was extracted by the ultrasonic -assisted enzymatic method .The ex-traction conditions were optimized through the single factor and orthogonal tests .The effects of the dosage of enzyme ,the concentration of extracting agent ,enzymolysis time ,temperature ,ultrasound time and the material/liquid ratio on pigment were studied in this paper .The optimum conditions were as follows :en-zyme dosage 4mg/g (petal weight) ,60% ethanol ,soaking for 80min at 50℃ with the material/liquid ratio of 1 :15 ,ultrasonic extraction of 15min .The stability experimental results showed that the pigment from magnolia liliiflora was applicable in acidic conditions .The NaCl and some carbohydrates had the effect of color-protecting .But the pigment was unstable against light ,alkaline environment and some metal ions such as Fe3+ and Cu2+ .
