摘要
目的通过检测表皮生长因子(EGF)相关蛋白在正常子宫内膜、增生性子宫内膜及子宫内膜癌组织中的表达,进一步探讨其临床意义。方法采用免疫组织化学法检测EGF、转换生长因子a(TGF-a)在正常子宫内膜、增生性子宫内膜及子宫内膜癌中的差异性表达;采用Real-time PCR方法检测上述因子的RNA水平。结果免疫组织化学结果示:EGF在正常子宫内膜组、增生性子宫内膜组及子宫内膜癌组中的阳性表达率分别为71.4%、72.2%和57.6%,子宫内膜癌组明显低于正常子宫内膜组及增生性子宫内膜组(均P<0.05);TGF-a在正常子宫内膜组、增生性子宫内膜组及子宫内膜癌组中的阳性表达率分别为42.9%、44.4%和66.7%,子宫内膜癌组的表达率明显高于正常子宫内膜组和增生性子宫内膜组(均P<0.05)。Real-time PCR方法检测正常子宫内膜组和子宫内膜癌组组织中EGF及TGF-a的表达,结果与免疫组织化学检测结果一致,EGF在子宫内膜癌中相对低表达,TGF-a呈相对高表达(均P<0.05)。结论 EGF及TGF-a的水平变化与子宫内膜癌的发病存在密切关系。
Objective To investigate the expression and clinical significance of epidermal growth factor (EGF) -related protein in normal endometrium, endometrial hyperplasia and endometrial carcinoma(EMC). Methods The protein and mRNA expression levels of EGF and transforming growth factor-a (TGF-a) in normal endometrium, hyperplastic endometrium and EMC were detected by using immunohistochemistry and real-time PCR, respectively. Results Immunohistochemistry showed that the positive expression rate was 71.4%, 72.2%and 57.6%for EGF and 42.9%, 44.4%and 66.7%for TGF-a in normal endometrium, hyperplastic endometrium and EMC, respectively. Compared with normal endometrium or hyperplastic endometrium, EGF expression significantly decreased and TGF -a expression obviously increased in EMC (all P&lt;0.05). These results were consistent with real-time PCR assay. Conclusion Changes in EGF and TGF-a expression are closely related to malignant potential of EMC.
