摘要
对枯草芽孢杆菌 (Bacillus subtilis) S2 1进行了摇瓶条件下的 D-核糖发酵条件的研究 ,主要考察了添加木糖、p H、溶氧水平以及采用补糖方式对发酵的影响 ,确定了 D-核糖发酵的最佳工艺条件 :D-木糖 5 0 g/L初始 p H7.0 ,初始葡萄糖浓度为 15 0 g/L ,碳酸钙 2 0 g/L ,并在发酵 36 h后 ,在装液量为 35 m L的 5 0 0 m L三角瓶中添加 0 .75 g/m L的葡萄糖溶液 2m L ,S2 1菌可积累 D-核糖达 5 1.1g/L。
Shake-flask culture conditions of D-ribose fermentation were studied with a Bacillus subtilis mutant S21.The paper investigated the factors affecting the yield of D-ribose,such as the amount of D-xylose in fermentation medium,pH、dissolved oxygen concentration and sugaring culture.It was suggested that the better fermentation conditon on laboratory scale was as following:D-xylose 50 g/L,initial pH7.0,initial glucose concentration 150 g/L,CaCO_320 g/L.After it cultured 36 h,2 mL 0.75 g/mL D-glucose solution should be added to the 500 mL flask contained 35 mL fermentation medium.Under such optimized conditions,the yield of D-ribose was about 51.1 g/L.
