摘要
目的 探讨SLE患者外周血B细胞中SLE活动相关的微小RNA(miRNA)表达与SLE活动时SLEDAI评分、补体C3水平的关系.方法 在芯片分析技术筛选出SLE患者外周血B细胞中SLE活动相关miRNA的基础上,利用茎环状引物实时定量-PCR技术在16例SLE重度活动期和16例稳定期患者外周血B细胞中进行验证.采用t检验和直线相关分析进行统计学分析.结果 茎环状引物实时定量-PCR技术验证发现SLE重度活动期5个miRNA(miR-191,miR-103,miR-17,miR-1224-3P,miR-1249(0.50±0.21,0.51±0.27,0.8±0.6,1.6±1.4,2.2±2.0)较稳定期(0.68±0.24,0.80±0.39,1.7±1.0,3.5±2.6,4.6±2.9)下调(t=-2.15,t=-2.26,t=-2.72,t=-2.45,t=-2.58,P均<0.05);miR-155(0.59±0.25)较稳定期(0.38±0.21)上调(t=2.36,P<0.05).同时血清补体C3下降程度与miR-191表达呈正相关(r=0.625,P<0.05),与miR-155表达呈负相关(r=-0.548,P<0.05).结论 SLE活动期与静止期患者外周血B细胞中存在着miRNA差异表达,且其中miR-191、miR-155与补体C3下降有关,提示miRNA可能参与SLE患者狼疮活动过程.
Objective To investigate the relationship between the disease activity related miRNAs in the peripheral blood B lymphocytes and SLEDAI score and Serum complement C3 level in systemic lupus erythmatosus (SLE) patients.Methods After identifying the miRNAs related to lupus activity by miRNA microarray chip analysis,Stem-loop real-time polymerase chain reaction (PCR) technology was used to confirm the related miRNAs in 16 active and 16 stable SLE patients.T-test and liner correlation correlation analysis were used for statistical analysis.Results Five down regulated miRNAs (miR-191,miR-103,miR-17,miR-1224-3P,miR-1249,0.50±0.21 vs 0.68±0.24,0.51±0.27 vs 0.80±0.39,0.8±0.6 vs 1.7±1.0,1.6±1.4 vs 3.5±2.6,2.2±2.0 vs 4.6±2.9; t=-2.15,t=-2.26,t=-2.72,t=-2.45,t=-2.58,P<0.05) and one upregulated miRNA (miR-155,0.59±0.25 vs 0.38±0.21; t=2.36,P<0.05) were found in severe active SLE patients by real-time PCR.The expression level of miR-191 was significantly positively correlated with serum C3 level (r=0.625,P<0.05),while miR-155 was significantly negatively correlated with serum C3 level (r=-0.548,P<0.05).Conclusion miRNAs are differentially expressed in peripheral blood B lymphocytes in severe active and stable SLE patients.miR-191 and miR-155 are significantly related to serum C3 levels,which indicate that miRNAs may play important roles in the status of lupus disease activity.
