摘要
目的:探讨整合素a5b1 mRNA表达水平对胃癌细胞黏附、迁移、髓外浸润过程的影响.方法:采用Western blot检测法及RT-PCR分别测量HTB-103、CRL-5822、CRL-5971及CRL-5973等不同对数生长期的白细胞株整合素a5b1蛋白表达水平及整合素a5b1 mRNA水平.将不同胃癌细胞株随机分为实验组及对照组,实验组加入整合素a5b1抗体,对照组加入同型IgG,观察两组胃癌细胞与ECV304细胞系的黏附率、细胞迁移率及白细胞穿过人工基质膜的浸润能力.结果:CRL-5822、CRL-5971及CRL-5973细胞整合素a5b1 mRNA相对表达量分别为(0.0821±0.0128)、(0.185±0.0082)、(0.798±0.042)明显高于HTB-103细胞(0.0002±0.0000)(P<0.05).观察组CRL-5822、CRL-5971及CRL-5973细胞株黏附率(52.16%±2.52%、44.22%±2.59%、33.58%±4.06%)、迁移率(45.96%±1.21%、38.86%±1.99%、24.65%±3.28%)及浸润率(29.85%±4.63%、21.31%±3.24%、13.67%±3.48%)、显著低于对照组黏附率(83.89%±7.21%、71.17%±7.38%、60.89%±10.59%)、迁移率(75.41%±9.51%65.78%±14.62%,49.91%±13.47%)及浸润率(42.63%±7.69%,29.92%±5.47%,21.59%±6.72%)(P<0.05),而两组HTB-103细胞黏附率、迁移率及浸润率无统计学差异(P>0.05).结论:黏附因子整合素a5b1可能参与胃癌细胞的形成、聚集、生长及浸润的过程,其表达水平与胃癌病程进展具有密切的关系.
AIM: To investigate the role of the adhesion molecule integrin a5bl in adhesion, migration and extramedullary infiltration of gastric cancer cells.METHODS: Immunohistochemistry and RT- PCR were used to measure integrin a5blprotein and mRNA expression levels in cell lines HTB-103, CRL-5822, CRL-5971 and CRL-5973. Different cell lines were randomly divided into an experimental group and a control group. The experimental group was incubated with anti-in- tegrin a5bl antibody, and the control group was incubated with control IgG. After incubation, the adhesion, migration and extramedullary infiltration of the above cells and ECV304 cells were assessed.
RESULTS: The relative mRNA expression levels of integrin a5bl were significantly higher in CRL-5822, CRL-5971 and CRL-5973 cells than in HTB-103 cells (0.0821 ± 0.0128, 0.185 ± 0.0082, 0.798 ± 0.042 vs 0.0002 ± 0.0000, P 〈 0.05). The rates of adhesion, migration and extramedul- lary infiltration in CRL-5822, CRL-5971 and CRL-5973 cells in the experimental groups were significantly lower than those in the control groups (adhesion (%): 52.16 ± 2.52 vs 83.89± 7.21, 44.22 ± 2.59 vs 71.17 ± 7.38, 33.58 ±4.06 vs 60.89 ± 10.59; migration (%): 45.96 ± 1.21 vs 75.41± 9.51, 38.86 ± 1.99 vs 65.78 ± 14.62, 24.65 ± 3.28 vs 49.91 ± 13.47; infiltration rate (%): 29.85 ± 4.63 vs 42.63 ± 7.69, 21.31± 3.24 vs 29.92 ± 5.47, 13.67 ± 3.48 vs 21.59 ± 6.72; P 〈 0.05 for all), but the above indexes did not differ in HTB-103 cells between the two groups (P 〉 0:05 for all). CONCLUSION: The adhesion molecule integrin a5bl may be involved in the adhesion, migration and extramedullary infiltration of gastric cancer cells.
