PPARγ C161→T、α-内收蛋白Gly460Trp基因多态性与原发性高血压的关系 被引量:3

摘要 目的探讨过氧化物酶体增殖物活化受体γ(PPARμ)C161→T及α-内收蛋白Gly460Trp基因多态性与原发性高血压(EH)的关系。方法原发性高血压患者(高血压组)262例与正常人群270例(健康对照组)为研究对象,应用聚合酶链反应—限制性片段长度多态性方法对PPARμ基因及α-内收蛋白Gly460Trp基因多态性位点进行分析,统计2种基因多态性频率并进行比较。结果高血压组PPARμC161→T基因型分布情况为TT型10例(3.82%),TC型70例(26.72%),CC型182例(69.46%),T型等位基因为90例次(17.18%),C型等位基区为434例次(82.82%);健康对照组PPAR_γC161→T基因型分布情况为TT型17例(6.30%),TC型119例(44.07%),CC型134例(49.63%),T型等位基因为153例次(28.33%),C型等位基因为387例次(71.67%),2组间分布比较差异有统计学意义(P<0.05)。高血压组α-内收蛋白Gly460Trp基因型分布情况为GlyGly型57例(21.76%),GlyTrp型为129例(49.23%),TrpTrp型为76例(29.01%),Gly型等位基因为243例次(46.37%),Trp型等位基因为281例次(53.63%);健康对照组α-内收蛋白Gly460Trp基因型分布情况为GlyGly型63例(23.33%),GlyTrp型为124例(45.93%),TrpTrp型为83例(30.74%),Gly型等位基因为250例次(46.30%),Trp型等位基因为290例次(53.70%),2组间分布比较差异无统计学意义(P>0.05)。结论中国人群中PPARγC161→T基因多态性与原发性高血压病相关;α-内收蛋白Gly460Trp基因多态性与原发性高血压无相关性。 Objective To investigate the relationship between peroxisome proliferator activated receptor gamma (PPARγ) C161 → T, α-adducin gene Gly460Trp polymorphism and essential hypertension (EH). Methods Patients with essential hypertension (hypertension group) were 262 cases, and 270 normal persons (control group) were enrolled in the research, using polymerase chain reaction restriction fragment length polymorphism of loci PPARγ gene and α-adducin gene Gly460Trp analysis, analyzed the 2 kinds of gene polymorphism frequency and compared between them. Results In hypertension group, PPAR3' C161 → T genotype distribution as follows: 10 cases of type TT (3. 82% ), type TC in 70 cases (26.72%), type C C in 182 cases (69.46%), the T allele was 90 cases (17.18 % ), C allele was 434 cases (82.82%) ; In healthy control group, distribution of PPARγ C161 → T gene type were TT type in 17 cases (6.30%), type TC in 119 ca- ses (44.07%), type CC in 134 cases (49.63%), the T allele was 153 cases (28.33%), C allele in 387 cases time (71.67%), compared the difference of distribution between the 2 groups showed statistically significant ( P 〈 0.05 ). Hypertension group alpha-adducin Gly460Trp genotype distribution as follows:GlyGly type in 57 cases (21.76%) , type GlyTrp in 129 cases (49.23%) , type TrpTrp in 76 cases (29.01%) , the Gly allele was 243 cases (46.37%) , the Trp allele was 281 cases ( 53. 63% ) ; healthy controls α-adducin Gly460Trp genotype distribution as follows: GlyGly type in 63 cases (23.33%), type GlyTrp in 124 cases (45: 93% ), type TrpTrp in 83 cases (30.74%), the Gly allele was 250 cases (46.30%), the Trp allele was 290 cases (53: 70% ), no statistically significant difference of distribution between the 2 groups were found ( P 〉 0.05). Conclusion Chinese population with PPARγ C161→T gene polymorphism correlated with essential hypertension; no correlation between α-adducin gene Gly460Trp polymorphism and essential hypertension.
