Determination of Puerarin in Kanggan Jiere Granule by HPLC

目的：利用HPLC法对抗感解热颗粒中葛根素的含量进行测定。方法：采用HPLC法,Diamonsil C18色谱柱（250 mm×4.6 mm,5μm）,流动相为甲醇-水（24∶76）,检测波长为250 nm,流速为1.0 mL/min。结果：葛根素在0.1841~1.841μg范围内呈良好的线性关系（r=0.9998）,RSD为1.65%（n=6）。葛根素平均回收率为99.32%。结论：本方法简便、可靠、准确,可用于抗感解热颗粒的质量控制。

Abstract： Objective： An HPLC-UV method was established to determine puerarin in Kanggan Jiere Granule. Methods ： Chromatographic separation was achieved on a Diamonsil Cla column （250 mm ×4.6 mm, 5 μm）. The mobile phase consisted of methanol and water （ 24 ： 76, v/v ） at a flow-rate of 1.0 mL ·min-1. The ultraviolet detector operated at 250 nm. Results： Linear calibration curves were obtained over the concentration range of 0.1841-1.841 μg · mL for puerarin （ r= 0.9998 ）, RSD=1.65% （ n=6 ）. The average recovery rate was 99.32% with RSD of 1.65%. Conclusion ： The method is simple, reliabe, accurate and it could be used in the determination of puerarin in Kanggan Jiere Granule.
