摘要
目的 建立测定血浆肝素 (Hp)浓度方法 ,探讨其临床应用可行性。方法 用微量发色底物法测定血浆 Hp浓度 ,监测了 1 8例肾病病人血透期间和 2例 DIC病人在用肝素治疗期间血浆肝素浓度。结果 标准曲线在 0 .1~ 0 .8u/ml范围内成线性关系。高、中、低三种浓度回收率分别为 90 .1 %、93.2 %和91 .4%。批内 CV=5.2 % ,批间 CV=7.1 % ,与 TCT法有良好的相关性 (r=0 .91 89,P<0 .0 1 )。肾病病人血透期间血液 Hp浓度为 0 .2 7~ 0 .61 u/ml,2例 DIC病人在皮下注射 Hp后 2 h和 3h分别达到 0 .36u/ml和 0 .33u/ml的峰值浓度。结论 此法简便、快速、特异、稳定、准确性好 ,成本低 ,可用于临床。
Objective To establish the method for assay plasma heparin(Hp) concentration and approach into the reliability of clinical application.Method Concentration of plasma Hp is determinated by micromethod chromogenic substrate and is oberserved in 18 hemodialysis patients suffering from in nephrosis and 2 DIC patients treated in Hp.Result The standard curve is linear in the 0 1~0 8u/ml range.The respective recovery rate are 90 1%,93 2%and 91 4% in hight,middle and low concentration.The within run CV is 5 2% and between run CV is 5 8%.There is a hight correlation with the result of the TCT( r =0 9189, P <0 01).Range of Hp concentration of hemodialysis patients is 0 27~0 61u/ml in a period of time of hemodialysis.The peak value of plasma Hp concentration of 2 DIC patients are 0 36u/ml and 0 33u/ml respectively for 2 and 3 hour after injecting Hp.Conclusion This method is simple,rapid,special,stable and accurate.The costs are reduced.It is suitable to clinical application.
