摘要
[目的]探索适用于浙江茄子品种"杭州红茄"的高效离体再生的途径和方法。[方法]以杭州红茄的子叶、下胚轴和带腋芽茎段为试材,研究了该品种从愈伤诱导、分化、增殖到生根壮苗过程的离体再生体系,对不同时期的培养基及激素配比进行筛选比较。[结果]愈伤组织的诱导培养基以MS+6-BA 2.0 mg/L+IAA 0.1 mg/L效果较好,分化的理想培养基为MS+ZT 0.2 mg/L+IAA 0.1 mg/L,而生根培养基以1/2MS+IAA0.1 mg/L较适宜。[结论]该研究获得了高效的杭州红茄离体再生方法,为其基因工程的开展提供了技术支持。
[Objective] To explore the efficient regeneration system of "Hangzhou Hongqie" which is the local cultivar of Solanum melongena L in Zhejiang.[Method] With the cotyledon,hypoeotyls and stem segments of "Hangzhou Hongqie" as explants,the regeneration system in vitro including callus induction,differentiation,proliferation and the rooting process was studied.The optimum mediums and hormone ratio in different periods were obtained.[Result] The results showed that the suitable medium for the callus induction was MS + ZT 1.0 mg/L + IAA 0.1 mg/L,and the ideal medium for differentiation was MS + 6-BA 2.5 mg/L + IAA 0.2 mg/L.On the other hand,the medium of 1/2MS + IBA 0.3 mg/L was better than MS for rooting.[Conclusion] High efficient regeneration system of "Hangzhou Hongqie" was established,which provide technical support for its genetic engineering.
