摘要
[目的]建立绶草高频再生体系。[方法]以野生绶草的花序轴为外植体,对外植体进行消毒和预培养后,以MS为基本培养基,添加不同浓度的6-BA筛选最适诱导培养基;以MS为基本培养基,添加不同浓度6-BA和IAA筛选最适增殖培养基;以1/2 MS为基本培养基,添加不同浓度NAA筛选最适生根培养基。[结果]最适诱导培养基为MS+6-BA2.0,最适增殖培养基为MS+6-BA1.5+IAA0.4,最适生根培养基为1/2MS+NAA1.5。[结论]该研究对绶草优良品种的改良和次级代谢产物研究有重要意义。
[Objective] To establish a high frequency regeneration system of Spiranthes sinensis.[Method] With rachis of wild Spiranthes sinensis as explant,disinfection and preculture was conducted.With MS as basic medium,adding different concentration 6-BA,the optimum induction medium was obtained; with MS as basic medium,adding different concentration 6-BA and IAA,the optimum proliferation medium was selected; with 1/2 MS as basic medium,add different concentration NAA for screen out the optimal rooting medium.[Result] The optimum medium for induction is MS + 6-BA 2.0 ; the optimum medium for proliferation is MS + 6-BA 1.5 + IAA 0.4 and 1/2 MS + NAA 1.5 is the best rooting medium.[Conclusion] The study has significance for improvement and secondary metatolites research of Spiranthes sinensis fine cultivar.
