红曲霉液体深层发酵生产红曲色素条件的研究 被引量:4

摘要 [目的]确定红曲霉产生红曲色素最适宜的培养基配方和培养条件。[方法]采用改变单一变量的方法,测定红色素色价的变化规律,确定红曲霉产生红色素最适宜的培养基配方和培养条件。[结果]试验确定了红曲霉产生红色素最适宜的培养基配方为:葡萄糖5%,酵母粉2%,CaCO30.2%,NaCl 0.1%,MgSO4·7H2O 0.1%,KH2PO40.2%,吐温20 0.05%;最适宜的培养条件为:pH 6.0,装液量为60 ml/250 ml三角瓶中接种10%(1/6)种龄为72 h的液体种子,30℃恒温摇床上振荡培养,转速为180 r/min,发酵6 d,红曲色素色价可达19.2 U/ml。[结论]该试验可为工业上大规模生产红曲红色素提供理论依据。 [Objective] To determine the optimal culture medium and fermentation conditions of Monascus pigment production by Monascus fumeus.[Method] By changing single variable,the variation law of red pigment was determined,the optimal culture medium formula was determined.[Result] The optimal culture medium was as follows:glucose 5%,yeast powder 2%,CaCO3 0.2%,NaCl 0.1%,MgSO4 · 7H2O 0.1%,KH2PO4 0.2%,Tween 20 0.05%.The suitable fermentation conditions were as follows:pH 6.0,broth volume of the shake flake 60 ml/250 ml,inoculation volume 10% (1/6),seed-age 72 h,culture temperature 30 ℃,rotation speed 180 r/rain,fermentation time 6 d,the Monascus pigment reached 19.2 U/ml.[Conclusion] The study can provide theoretical basis for the large scale production of Monascus pigment.
