摘要
目的观察三维适形调强放疗(IMRT)对鼠骨细粒棘球蚴生发细胞生长及凋亡的影响,探讨三维适形调强放疗治疗骨棘球蚴病的临床效果。方法选取骨细粒棘球蚴病大鼠,将骨细粒棘球蚴内囊中的生发细胞进行分离,并进行体外培养,将体外培养成功的骨细粒棘球蚴的生发细胞分为IMRT放疗组及空白对照组。IMRT放疗组给予总剂量为40Gy的IMRT放疗,空白对照组只给予假照射作为阴性对照。于放疗结束24h后,采用MTT法检测IMRT对骨细粒棘球蚴生发细胞生长的抑制作用,Hoechst 33258荧光染色观察IMRT对骨细粒棘球蚴生发细胞凋亡的影响,观察IMRT对鼠骨细粒棘球蚴病的治疗效果。结果 IMRT作用24h后,MTT法检测结果可观察到,IMRT放疗组的骨细粒棘球蚴生发细胞吸光度(OD值=0.213±0.070),远低于空白对照组的(0.792±0.098),差异有统计学意义(P<0.05),存在明显的增殖抑制现象;Hoechst 33258荧光染色可观察到IMRT治疗组的骨细粒棘球蚴生发细胞呈致密浓染的强蓝色荧光,存在明显的核固缩、染色质凝集等细胞凋亡现象,而空白对照组未出现。结论 IMRT对骨细粒棘球蚴生发细胞生长具有明显的抑制作用并可诱导生发细胞凋亡。
Objective To observe the possibility of the growing influence and apoptosis of germinal cells of intensity modulated radiation therapy (IMRT) to the protoscolex of bone eehinoeoceosis in rats,and to investigate the clinical effect of intensity modulated radiation therapy on bone hydatid disease. Methods Purebred bone echinococcosis rats were selected and the germinal cells of bone echinococcosis was separated,and cultured in vitro. The germinal cells of bone echinococeosis was divided into IMRT radiotherapy group and blank control group. IMRT radiotherapy group, giving a total dose of 40 Gy IMRT radiotherapy, blank control group only blank irradiation as a negative control. In radiation therapy after 24 h. And determined by MTT method was used to detect IMRT for growing influence of germinal cells growth inhibition, Hoechst 33258 fluorescent staining observation of IMRT for germinal cells apoptosis, in order to observe the clinical effect of intensity modulated radiation therapy on bone hydatid disease. Results 24 hours later, The proliferation of germinal cells of bone echinococcosis cells was significantly inhibited by MTT assay in the IMRT group. Distinct morphologic changes of cell apoptosis such as karyopyknosis and chromatic agglutination were observed by Hoechst 33258 staining. Conclusion It is evident that the IMRT can promote the growing influence and apoptosis of germinal cells of bone echinococcosis.
