摘要
目的:研究TGF-β1/Smad2信号通路在野百合碱诱导的肺动脉高压大鼠中的表达。方法:18只雄性SD大鼠随机分为对照组和模型组,采用皮下注射野百合碱(60 mg/kg)的方法复制肺动脉高压模型。野百合碱注射5w后,测定大鼠右心室收缩压(RVSP)和右心室肥厚指数(RVHI)。Western blot检测肺组织中TGF-β1、p-Smad2和Smad2的表达水平。结果:模型组大鼠的RVSP、RVHI明显高于对照组,模型组大鼠肺组织中TGF-β1、p-Smad2的表达明显高于对照组。结论:TGF-β1/Smad2信号通路可能参与了肺动脉高压发病过程。
Objective:To explore the expression of TGF-IM/Smad2 signaling pathway in monocrotaline induced pulmonary hypertension in rats. Methods: 18 male Sprague-Dawley rats were randomly divided into control group and model group. The rats in the model group were underwent a subcutaneous injection of 60 mg/ kg monocrotaline(MCT). Five weeks after injection, fight ventricular systolic pressure (RVSP) and right ventficular hypertrophy index (RVHI) were measured. The expression levels of TGF-β1, p-Smad2 and Smad2 in the lung tissue were detected by Western blot. Results: RVSP and RVHI in the model group were significantly higher than that in the control group. The expression of TGF-β1 and p-Smad2 in the lung was also significantly higher than that in the control group. Conclusion: TGF-β1/Smad2 signaling pathway may be involved in the pathogenesis of pulmonary hypertension.
