摘要
目的:探讨姜黄素对人牙髓细胞(human dental pulp cells,HDPCs)细胞周期及凋亡的影响。方法:体外培养人健康牙髓细胞,不同浓度姜黄素(curcumin)处理细胞后流式细胞术检测细胞周期,TUNEL染色法观察细胞凋亡,实时荧光定量PCR及免疫蛋白印迹分别检测p300 mRNA及蛋白水平。结果:流式细胞术及TUNEL结果显示姜黄素处理人牙髓细胞后,G0/G1期细胞比例上升,S期细胞比例下降,凋亡细胞增加;实时荧光定量PCR及免疫蛋白印迹结果显示姜黄素抑制p300 mRNA及蛋白的表达。结论:姜黄素使细胞周期停滞在G0/G1期,诱导牙髓细胞凋亡,其机制可能是通过下调p300活性。
Objectives: The purpose of this study was to investigate the role ofcurcumin in the cell cycle and apoptosis of HDPCs. Methods: The cell cycle and apoptosis of the HDPCs were analyzed using flow cytometry and TUNEL staining, respectively. Real time-qPCR and western blotting analysis were utilized to analyze the mRNA and protein expression of p300 after curcumin impacted the HDPCs. Results: Curcumin arrested cells in the G0/G1 phase and triggered apoptosis of HDPCs. p300 was down-regulated at the mRNA and protein levels by curcumin. Conclusion: Curcumin affected the the cell cycle and apoptosis of HDPCs by down-regulation of p300.
