摘要
报道了一种快速分离纯化 Gluconbacter melanogenus的方法。通过优化培养基成分和培养温度并改变分离操作技术 ,可以在短时间内使菌株在分离平板上出现清晰的溶钙圈 ,根据溶钙圈的大小 ,可以快速筛选到优良菌株。将溶钙圈直径大小不同的菌株斜面培养好后 ,通过 1 2代小三角瓶液体传代 ,并与生产岗位现用菌株比较 ,证明溶钙圈大的菌株发酵率确有提高。
An effective method of isolation and purification of Gluconobacter melanogenus is reported.By optimizing medium compositions,culture temperature and improving isolation technique,the dissolved calcum can be seen clearly and rapidly.According its diameter,excellent strains are selected.Different diameter stains are inoculated liquid to liquid in flasks for twelve generations,and compare with the strain use in production.It is proved that the big dissolved calcium strains have higher fermentation rate.
