平菇提取物对激素性股骨头坏死大鼠模型的药效作用研究 被引量:5

摘要 [目的]评价平菇(侧耳)水提物对大鼠激素性股骨头坏死的药效作用。[方法]采用大肠杆菌内毒素联合甲基强的松龙制备大鼠激素性股骨头坏死模型,实验设正常组,模型组,阿仑膦酸钠组和侧耳水提物组。通过行为学研究观察大鼠一般状态、抓力、自主活动和悬尾时间。观察6W后处死大鼠,取出连同大、小转子在内的大鼠双侧股骨头用HE染色进行组织病理学观察。[结果]模型组大鼠出现畏寒倦卧,消瘦,食欲下降,精神萎靡,活动迟缓,对外界影响反应差等肾阳虚表现;组织病理学显示其股骨头髓腔呈现大量脂肪细胞,并伴有骨小梁狭窄和空骨陷窝增加等典型骨坏死症状。经平菇水提物治疗后大鼠食欲恢复正常,偶有消瘦,行动活泼,精神状态良好。给药后大鼠抓力显著升高,自主活动次数明显增加,悬尾时间显著延长。病理学观察发现平菇水提物能显著改善股骨头病变,抑制其坏死进程。而阳性药阿仑膦酸钠对激素性股骨头坏死大鼠行为学指标没有明显作用,对组织病变有轻微改善作用。[结论]平菇水提物可显著改善激素性股骨头坏死大鼠行为学指标并抑制股骨头组织坏死进程,效果优于阿仑磷酸盐。 [Objective]To evaluate the therapeutic effect of aqueous extracts of Pleurotus ostratus on steroid-induced femora] head necrosis(SFHN).[Methods] The models of rat SFHN were induced by endotoxin combined with methylprednisolone.80 Wistar rats were randomized into the normal group,model group,the alendronate group and aqueous extracts from Pleurotus ostratus group.The effects were detected by observing general state,measuring grip strength,autonomic mobility frequency and tail suspension time.The changes of the femoral head organization were observed by hematoxylin-eosin staining,[Results] The model rats performed the similar symptoms of kidney-yang,such as loss of appetite,listlessness,weight loss,slow movements,poor response to outside influences.After treatment of aqueous extracts from Pleurotus ostratus,the rats' appetite improved,moved lively,and in good spirits,though lost weight at times.After administration,the grip strength of the rats was significantly enhanced.Autonomic mobility frequency was remarkably increased.And the tail suspension time was significantly prolonged.The pathology revealed that aqueous extracts from Pleurotus ostratus can significantly improve the femoral head cartilage surface depression,and reduce the degree of degeneration and necrosis of chondrocytes.Alendronate did not reveal any improvement in rats' SFHN behavior and the femoral head tissue pathological changed.[Conclusion] Aqueous extracts from Pleurotus ostratus significantly improved the indexes of the SFHN in rat behavior and a significant reversal of tissue necrosis of femoral head.
