摘要
[目的]观察夹脊电针干预脊髓损伤大鼠髓鞘再生和运动功能恢复作用。[方法]108只SD(Sprague Dawley)雄性大鼠随机分为假手术组、模型组、夹脊电针治疗组(EA组),每组再分为术后1天、7天、14天三个时间点组(n=12)。假手术组只切除椎板,其余两组建立脊髓损伤模型后模型组不予治疗,EA组取损伤周围夹脊穴进行电针治疗(疏密波,2/100HZ,1mA),每次20min,每天一次。各组大鼠于各时间点进行BBB(Basso,Beattie,Bresnahan)运动功能评分,应用LFB(Luxol Fast Blue)法进行髓鞘染色,western blot方法检测髓磷脂碱性蛋白(Myelin Basic Protein,MBP)的表达。[结果]术后1天EA组与模型组BBB评分无明显差异(P=0.724),7天和14天时EA组大鼠BBB评分逐渐增高,与模型组比具有显著性差异(P<0.01);LFB髓鞘染色发现脊髓损伤后白质区域有较大面积的染色变浅,间隙变稀疏、排列不均匀,且有大量空泡存在,术后1天模型组大鼠髓鞘平均积分光密度值(Integrated Optical Density,IOD)较假手术组明显降低(P<0.01),EA组较模型组无明显差异(P=0.945);7天和14天EA组髓鞘含量显著增多,与模型组比较具有显著性差异(P<0.05或P<0.01);术后EA组MBP蛋白相对表达逐渐增多,在各时间点较模型组均具有显著性差异(P<0.01)。[结论]夹脊电针可能通过增加MBP的表达,有效促进脊髓损伤后的轴突再髓鞘化,从而进一步促进大鼠神经功能的恢复。
[Objective] To investigate the intervention ofJia-Ji EA on myelin sheath regeneration and motor functional recovery of spinal cord injury in rats.[Methods] 108 adult male SD rats were randomized into the sham control group(sham group),the model control group(modal group),Jia-Ji EA group(EA group),and each group was further divided into day 1,day 7,day 14 post-operation(n=12).After spinal cord injury models were established,EA(disperse-dense wave,2/100Hz,lmA) was applied to acupoints around the damaged segment once a day for 20 mir.After BBB locomotor rating scale was rsed at each time point,rats were executed and then Luxol Fast Blue(LFB) was used for myelin staining and Western blot was used to detect the expression of myelin basic protein(MBP).[Results] Day 1 post-SCI,there were no statistical differences of BBB scores between EA group and model group(P=-0.724).Day 7 and 14 post-SCI,BBB scores increased gradually in EA group,and there was a significant difference compared with model group (P〈0.01).LFB myelin staining showed that color of largearea in white matter became shallow,interval became sparse with amount of cavities and arrangement became irregular.Day 1 post-SCI,IOD values in model group were significantly decreased compared with the sham control group(P〈0.01),but no significant difference compared with EA group(P=0.945).Day 7 and 14 post-SC1,the number of myelin sheath gradually increased in EA group and there was a significant difference compared with model group(P〈0.05 or P〈0.01).In EA group,the expression of MBP increased gradually and the differences in each time point were all significant compared with the model control group(P〈0.01).[Conclusions] EA can effectively promote functional recovery after SCI probably by means of increasing the expression of MBP and myelin sheath regeneration.
