酶解谷壳制备阿魏酸的工艺优化 被引量:2

摘要 目的:利用响应面优化谷壳中阿魏酸的提取工艺。方法:采用木聚糖酶酶解谷壳提取阿魏酸,采用HPLC法对阿魏酸提取率进行测定。在单因素试验基础上,利用Design Expert数据处理软件进行响应面实验设计,确定最佳提取工艺。结果:木聚糖酶可有效降解谷壳得到阿魏酸。响应面优化得到的最佳提取工艺为加酶量1.2%、酶解时间31min、酶解温度48℃,酶解pH4.8,该条件下阿魏酸提取率为8.04mg/g结论:利用响应面分析方法对谷壳中阿魏酸的提取工艺进行优化,可获得最佳工艺参数。 Objective: To find optimal process conditions for the solvent extraction of ferulic acid from chaff by response surface methodology. Method: The Xylanase was used to prepare ferulic acid from chaff. And the extraction ratio of ferulic acid was obtained by HPLC. On the basis of single factor experiment, using the Design Expert data processing software to carry on the response surface design of experiment. Result: Xylanase can degrade chaff effectively to get ferulic acid. The optimum processes are: xylanase concentration is 1.2%, extraction time is 31min, extraction temperature is 48 ℃, and extraction pH is 4. 8. Conclusion : Using the response surface analysis method to optimize the extraction process of fenalic acid and optimum process parameters can be obtained.
