超临界二氧化碳萃取糖蜜酒精废液中黄酮类化合物的工艺研究 被引量:2

摘要 研究了CO2超临界流体萃取糖蜜酒精废液中黄酮类化合物的影响因素,在单因素试验的基础上,以总黄酮提取率为主要评价指标,进行了L9(34)正交优化。结果表明,通过单因素实验,选择甲醇为夹带剂,确定萃取体系的温度、压力、时间及夹带剂流量为正交实验的主因素和取值范围。正交实验得最佳工艺条件为:萃取体系温度50℃、萃取时间120min、萃取压力300Bar(1Bar=0.1MPa)、甲醇夹带剂流量0.3mL/min,此条件下试验超临界二氧化碳萃取的黄酮类化合物的总量为0.289%,对DPPH自由基清除率为85.2%,确定了其中一种物质为4',5,7-三羟基-3',5'-二甲氧基异黄酮。由方差分析可知温度对萃取黄酮类化合物总量的影响达极显著水平,各因素影响顺序为萃取温度>萃取时间>萃取压力>夹带剂用量。 The effects of the extraction parameters on the yield of flavonoids from molasses vinasse were studied. The process parameters were optimized by L^9 (3^4 ) orthogonal tests based on single factor tests with the yield of total flavonoids as index. The optimal process parameters were: 50℃, 300 Bar ( 1Bar =0. 1MPa), 120min and 0. 3mL/min. The maximum extraction yield of flavonoids compounds was 0. 289%. The flavonoids had 85. 2% scavenging effect on DP- PH. One of the flavonoid compounds was identified as 5, 7, 4'- trihydroxy - 3', 5'- dimethoxyflavone. The results showed that the order of the extraction parameters affecting the yield of flavonoids is as follows : temperature 〉 time 〉 pressure 〉 cosolvent amount.
