草乌与瓜蒌、白及配伍对心肌细胞毒性与增毒机制研究 被引量:3

摘要 目的:探讨草乌、草乌与瓜蒌和草乌与白及配伍对大鼠心肌细胞毒性作用与增毒作用机制。方法:体外培养大鼠心肌细胞,给予2%,8%,16%浓度的含药大鼠血清作用于心肌细胞后,采用MTT法测定心肌细胞活力,比较对心肌细胞毒性大小,再利用试剂盒测定细胞外乳酸脱氢酶(LDH)含量、Na+-K+-ATP酶活力与含量以及细胞凋亡率,研究增毒机制。结果:与对照组相比,随着大鼠含药血清浓度的升高,心肌细胞存活率下降(P<0.05)。给药组LDH含量显著升高(P<0.05);Na+-K+-ATP酶活力随着浓度升高而降低(P<0.05);Na+-K+-ATP酶含量明显降低(P<0.05),血清浓度在8%,16%时,细胞凋亡率明显升高(P<0.05)。结论:草乌对心肌细胞有明显毒害作用,与瓜蒌、白及配伍后毒性增强;其增毒的机制是能显著增大心肌细胞膜通透性,破坏细胞能量系统,诱导细胞自身凋亡,加速细胞死亡。 Objective:To study the toxic effect and mechanism on rat&#39;s myocardial cell of Radix Aconiti Kusnezoffii and the Compatibility of Radix Aconiti Kusnezoffii with Fructus Trichosanthis ,Bletillae Rhizoma .Methods:based on vitro myocardial cell culture , it was given with serum with 2%,8%,16%drugs,dertermined myocardial cell vitality by MTT for comparing the toxicity to myocardial cell . Then determine the content of LDH ,vitality of Na +-K+-ATP enzyme,content of Na +-K+-ATP enzyme,cell apoptosis rate by respectively kits to research the mechanism .Results:compared with control group , with the content of drugs in serum rise , myocardial cell vitality decline(P〈0.05);the content of LDH in given group rise obviously (P〈0.05);the vitality of Na +-K+-ATP enzyme of given groups decline (P〈0.05);the content of Na +-K+-ATP enzyme decline obviously (P〈0.05);cell apoptosis rate of given groups rise obviously as serum with 8%,16%drugs (P〈0.05).Conclusion:Aconiti Kusnezoffii Radix have strong harm for cell , and after compatibility with anti-drug, toxicity is more stronger than itself .The toxicity increasing mechanism is knowed that Aconiti Kusnezoffii Radix can enlarge permeability of myocardial cytomembrane , destroy energy system of cell , induce cell apoptotic , which finally accelerate the death of myocardial cell .
