摘要
目的:观察针刺对AD模型大鼠海马凋亡相关蛋白Caspase-3的影响。方法:SD大鼠20只,连续6w腹腔注射D-半乳糖(120mg·kg-1)和亚硝酸钠(45mg·kg-1)建立AD模型后随机分成模型组、针刺组(n=10),同时设立非模型组(n=10)。造模后第21d,针刺组大鼠予以针刺百会和肾俞穴治疗,而模型组大鼠不予以治疗。针刺治疗结束后大鼠立即处死,采用免疫组化法检测非模型组、模型组、针刺组大鼠海马Caspase-3表达。结果:免疫组化结果显示模型组大鼠海马Caspase-3表达明显增加,与对照组比较有显著差异(P<0.01);而针刺组Caspase-3的表达明显减少,与模型组比较(P<0.01)。结论:针刺可抑制大鼠海马Caspase-3表达。
Objective:To observe the effect of acupuncture on apoptosis associated protein Caspase-3in hippocampus of AD rat.Methods:Thirty SD rats were randomly divided into control group,AD model group and acupuncture group(n=10).The rats AD model group and acupuncture group were continuous intraperitoneal injection of D-galactose(120mg·kg^-1)and sodium nitrite(45mg·kg^-1)for 6weeks,and the rats of control group were injected saline instead.21dafter modeling,the rats of acupuncture group treated with acupuncture of Baihui,Shenshu acupoint therapy.Rats in model group were not treated.Rats were sacrificed immediately after the acupuncture treatment.Caspase-3expression in hippocampus of rat was analyzed by immunohistochemical.Results:The immunohistochemistry results showed that expression of Caspase-3in hippocampus of rats in model group increased obviously,compared with the control group(P〈0.01),The expression of Caspase-3in the acupuncture group decreased significantly,compared with the model group(P〈0.01).Conclusions:Acupuncture can inhibit the expression of Caspase-3in hippocampus of AD rat.
