青蒿素对糖尿病大鼠肾组织NF-κB的DNA结合活性升高的抑制作用 被引量:3

摘要 目的探讨青蒿素对实验性糖尿病大鼠肾组织核因子-κB(NF-κB)的DNA结合活性升高的抑制作用。方法选择雄性SD大鼠15只,随机分成正常对照组(A组)、糖尿病组(B组)及青蒿素治疗组(C组),采用腹腔单剂量注射链脲佐菌素(STZ)65 mg/kg方法建立糖尿病大鼠模型。C组给予青蒿素300 mg/(kg·d)腹腔注射。于实验第4周应用电泳迁移率改变分析(EMSA)法检测3组大鼠肾皮质细胞NF-κB的DNA结合活性,并观察实验大鼠肾脏的病理学变化。结果 B组大鼠肾细胞NF-κB的DNA结合活性显著上调;C组NF-κB的DNA结合活性显著较B组下降,肾脏组织的病理学损伤亦减轻。结论青蒿素可明显改善糖尿病大鼠肾脏组织病理学改变,通过抑制糖尿病大鼠肾脏组织细胞NF-κB的DNA结合活性上调可能是相关作用机制之一。 Objective It is to investigate the inhibitory effect of artemisinin on the upregulation of DNA-binding activity of NF-κB in kidney tissue of experimental diabetic rats.Methods 15 male SD rats were selected and randomly divided into 3 groups:normal control group(A group),diabetic group(B group),artemisinin treatment group(C group).The diabetic rat models were established by intraperitoneal injection with STZ 65 mg/kg.Group C was given artemisinin 300 mg/(kg · d) by intraperitoneal injection.The DNA-binding activity of NF-κB in the kidney cortex cells was detected by EMSA,while renal pathologic changes were observed after 4 weeks' treatment.Results The DNA-binding activity of NF-κB was significantly activated in renal cortices in group B,and the activity was partly reversed in group C.Accordingly,renal pathologic changes in group C were significantly improved.Conclusion Artemisinin can obviously improve the pathologic changes of renal tissue in diabetic rats,one of the correlated mechanism may partly through inhibiting the DNA-binding activity of NF-κB in renal cortices.
