肝脏营养物质代谢的仔猪肝-门静脉血插管技术的建立 被引量:6

Establishment of Techniques for Implanting Hepatic-Portal Catheters in Piglets for Studying Hepato-Nutrients Metabolism

摘要 本试验目的是在仔猪上建立肝静脉-门静脉-肠系膜静脉-颈动脉血管插管系统,为仔猪肝脏营养物质的代谢研究提供技术支持。选择健康、体重(20.0±1.1)kg的杜×长×大三元杂交去势公仔猪18头,分别在肝静脉、门静脉、肠系膜静脉和颈动脉安装血管插管,待仔猪恢复正常采食量5 d后,通过肠系膜静脉连续灌注1%对氨基马尿酸,采集采食前0.5 h和采食后0.5、1.5、3.0、5.0、7.5 h肝静脉、门静脉和颈动脉的血液,测定仔猪肝静脉和门静脉的血流速度。结果显示,本试验安装的导管系统保证血管疏通性良好的平均时间为16 d,采食后门静脉和肝静脉的血流速度上升,门静脉流速最高为2.80 L/(kg·h)和981.40 mL/min;肝静脉流速达到4.04 L/(kg·h)和1 414.65 mL/min。18头试猪中的12头可成功用于肝脏营养物质代谢研究,肝静脉和门静脉的血流变化趋势相似。 This study was aimed to establish techniques for implanting arterial and venous catheters in piglets to provide technical support for studying hepato-nutrients metabolism. A total of 18 healthy Duroc×Landrace×Yarkshire castrated piglets weighted (20.0±1.1) kg were selected, and hepatic vein, portal vein, mesenteric vein, carotid artery catheters were implanted respectively. After 5 days recovery period, a 1% solution of p-amino hippurate (pAH) was infused continuously into the mesenteric catheters. The blood samples from hepatic vein, portal vein and carotid artery were collected at 0.5 h before feeding and at 0.5, 1.5, 3.0, 5.0 and 7.5 h after feeding, respectively. Hepatic vein and portal vein blood flow were detected. The results showed that the average time of keeping unobstructed in blood vessels of the implanted catheter system was 16 days. Hepatic vein and portal vein blood flow velocity increased after feeding. Portal vein flowe reached 2.80 L/(kg·h) and 981.40 mL/min. Hepatic vein flow reached 4.04 L/(kg·h) and 1 414.65 mL/min. Twelve of the eighteen test piglets can be successfully used for studying hepato-nutrients metabolism. Hepatic vein and portal vein blood flow tends to similarly change.
