银杏叶中黄酮类物质的酶法提取研究 被引量:15

摘要 以当地丰富的银杏叶为原料,采用纤维素酶辅助提取法对银杏叶中主要活性物质黄酮类物质进行浸提,通过单因素试验和正交试验,研究不同条件对提取率的影响,结果表明:酶解温度为45℃、酶处理pH为4、酶用量为0.3 mg/mL、酶解时间为2 h,在此条件下,银杏叶总黄酮提取得率为2.80%,与未添加纤维素酶相比,总黄酮得率增加了45.83%。在此基础上,通过比较银杏叶提取物和黄酮苷元对照品的高效液相图谱得出,银杏叶提取物中黄酮类物质主要是槲皮素、山奈酚、异鼠李素。 The paper took Taizhou Ginkgo Biloba leaves as raw material,using the extraction technology of the flavonoids from Ginkgo Biloba leaves by Cellulase.By experiments of the single factor and orthogonal analysis methodology to study the impact of different extraction conditions on the extraction yield, the results showed that the temperature of enzymatic hydrolysis was 45 ℃, pH =4, consumption of Cellulase was 0.3 mg/mL,the time of enzymatic hydrolysis was 2 h. On this condition, the extraction yield of flavonoids from Ginkgo biloba leaves was2.80 %,Which increased 45.83 %, compared with the non-using cellulase-assistant technology. Based on this, by comparing with the standard substance of HPLC-ESIMS chromatogram of the extract of Ginkgo Biloba leaves, the major flavone glycosides of flavonoids from Ginkgo Biloba leaves were quercetin, keampferol, isorhamnetin.
