摘要
目的观察兔脂肪间充质干细胞移植治疗眼表损伤的疗效。方法体外培养获得兔脂肪间充质干细胞,以羊膜为载体将其移植于兔角膜碱烧伤模型,以单纯羊膜移植为对照。术后1、2、4、8周进行角膜混浊程度、荧光素染色、新生血管形成情况的评分,计算新生血管面积。结果除术后1周、2周角膜混浊程度评分及术后1周荧光素染色评分两组之间无差异外,其它各检测时点实验组角膜混浊程度、新生血管形成及荧光素染色评分在治疗前后的变化量均大于对照组,差异均有统计学意义(均P<0.05)。实验组角膜新生血管面积在各检测时点均小于对照组,差异均有统计学意义(均P<0.05)。结论兔脂肪间充质干细胞移植治疗碱烧伤模型的眼表损伤有较好疗效。
Objective To examine the clinical efficacy of rabbit adipose derived mesenchymal stem cells transplantation for ocular surface damage. Methods The rabbit adipose derived mesenchymal stem cells were cultured,and then transplanted to the rabbit corneal alkali burn model with the amniotie membrane as a carrier. In the control group, the amniotic membrane was transplanted only. Scoring for corneal opacity, fluoreseein staining and neovaseularization was conducted,and the area of newly- formed blood vessels measured 1,2,4 and 8 weeks after operation. Results The corneal opacity, fluorescein staining and neovaseularization were significantly improved in the cell treatment group when compared with the control group. Significant differences were noted in these variables between the two groups at each measured time point except 1 and 2 weeks after operation for corneal opacity and 1 week after operation for fluorescein staining (P〈0.05). The area of new blood vessels was much smaller in the cell treatment group than in the control group with difference being significant (P〈0.05). Conclusion The rabbit adipose-derived mesenchymal stem cells transplantation is effective for the treatment of the ocular surface burn following alkali in jury.
