摘要
目的:研究原花青素(PC)对体外阿尔茨海默病模型(AD)的预防作用。方法:H2O2100μmol/L诱导PC12细胞损伤18 h复制体外阿尔茨海默病模型,原花青素80 mg/L、40 mg/L、20 mg/L、Tempol 800μmol/L于造模前30 min预处理后,双抗体一步夹心法酶联吸附实验(ELISA)检测细胞上清液中硫氧还蛋白(Trx)含量;黄嘌呤氧化酶法检测细胞中的总超氧化物歧化酶(T-SOD)活力;硫代巴比妥酸盐法测定细胞上清液的丙二醛(MDA)。结果:H2O2100μmol/L诱导PC12细胞损伤后,细胞内Trx浓度和T-SOD活力显著降低,MDA含量显著升高(P<0.01);原花青素各剂量组可增高Trx浓度和T-SOD活力,降低MDA含量(P<0.01,P<0.05)。结论:原花青素能减轻H2O2引起PC12细胞的氧化应激损伤。
Objective: To investigate the preventive effects of proanthocyanidins( PC) on Alzheimer's disease in vitro. Methods: Alzheimer's disease was mimicked in vitro by PC12 cell line injury induced with H2O2( terminal concentration,100 μmol/L) after incubation for18 h. The cell lines were pre-treated with PC in dose of 80 mg /L,40 mg /L and 20 mg /L as well as Tempol( 800 μmol /L,positive control) 30 min before modeling. ELISA was performed to detect the content of Trx in the cellular supernatant. Xanthine oxidase method was used to determine the T-SOD activity of cells,and thiobarbituric acid was used for measuring the content of MDA. Results: After induction of the PC12 cell injury with H2O2in dose of 100 μmol /L,the cellular Trx content and T-SOD activity were markedly decreased,whereas the content of MDA was increased significantly( P〈0. 01). However,treatment with PC in different dose had increased the content of Trx and boosted T-SOD activity,yet reduced the content of MDA in PC12 cell lines( P〈0. 01,P〈0. 05). Conclusion: Proanthocyanidins may alleviate the oxidative damage in PC12 cells induced with H2O2.
