摘要
目的研究银屑病患者外周血T细胞主控基因RUNX1及其靶基因SLC9A3R1、人类白细胞抗原(HLA)-C及磷酸激酶(PKC)-βⅠ的mRNA表达,进一步了解RUNX1及其下游基因与银屑病外周血T细胞活性异常的关系。方法取20例银屑病患者及20名健康对照外周血,分离、培养、扩增外周血T细胞并鉴定纯度;提取纯化mRNA并反转录合成cDNA,实时荧光定量聚合酶链反应(RFQ-PCR)检测RUNX1、SLC9A3R1、HLA-C及PKC-βⅠ在mRNA水平表达。结果银屑病患者外周血T细胞RUNX1、SLC9A3R1、HLA-C及PKC-βⅠmRNA表达水平均高于健康对照组,银屑病组RUNX1、SLC9A3R1、HLA-C及PKC-βⅠmRNA与对照组的表达水平比值分别为1.807 1、2.249 4、2.074 4及2.378 4。结论 RUNX1及其靶基因表达水平的增高可能引起外周血T细胞的活化,并参与银屑病的发病。
Objective To assess the mRNA expression of transcription factor RUNX1 and its target genes SLC9A3R1, HLA-C and PKC-βⅠin the peripheral blood T cells derived from patients with psoriasis, which may pro-vide insights into the aberrant relationship between RUNX1 downstream gene expression and peripheral blood T cells. Methods Peripheral blood samples were collected from 20 patients with psoriasis vulgaris and healthy subjects, fol-lowed by isolation, culture, amplification and purification of peripheral blood T cells. The mRNA was extracted from the T cells and reversed to cDNA. Real time fluorescent quantitative PCR was performed to determine the expressions of RUNX1, SLC9A3R1, HLA-C and PKC-βⅠmRNA. Results Compared with healthy subjects, the expression levels of RUNX1, SLC9A3R1, HLA-C and PKC-βⅠmRNA were 1.807 1, 2.249 4, 2.074 4 and 2.378 4 fold higher in pa-tients with psoriasis. Conclusion Heightened expression of RUNX1 and its target genes may have participated in the activation of peripheral blood T cells which are associated with the pathogenesis of psoriasis.
