摘要
利用猪胰弹性蛋白酶和尿激酶限制性酶切牛纤溶酶原 ,获得了纤溶酶原Kringles结构域片段Kringles1 -3 (K1 -3 )和Kringles1 -4 (K1 -4 ) .研究发现 ,在没有自由巯基供体存在的条件下 ,尿激酶酶切牛纤溶酶原也能够产生K1 -4 ,从而认为存在由尿激酶酶切牛纤溶酶原直接产生K1 -4的途径 .所获得的K1 -3和K1 -4对恒河猴脉络膜
Kringle 1-3 (K1-3) and Kringle 1-4 (K1-4) of natural bovine plasminogen were obtained through plaminogen proteolysis by elastase and urokinase. Without the participant of free disulfhydryl donor L-Cys, urokinase can also hydrolyses plasminogen to generate K1-4, thus it was considered there may be another path way of K1-4 generation, in which urokinase hydrolyse plasminogen to generate K1-4 directly. The K1-3 and K1-4 we acquired exhibited rhesus choroid-retinal endothelial cell proliferation inhibition activity in a dose dependent way.
